祛瘀化浊方对非酒精性脂肪肝模型大鼠脂代谢及肝功能的影响 被引量:8

Effects of decoction for resolving stasis and turbidity on lipid metabolism and liver function of rat models with non-alcoholic fatty liver

摘要 目的观察祛瘀化浊方对非酒精性脂肪性肝病(NAFLD)模型大鼠脂代谢和肝功能的影响,探讨该方治疗NAFLD的机制。方法将80只成年雄性SD大鼠随机分为正常组、模型组、多烯磷脂酰组、祛瘀化浊汤组,每组20只。除正常组外,其余组均采用高脂饲料喂养84 d制作NAFDL模型。造模成功后,多烯磷脂酰组给予多烯磷脂酰胶囊水溶液灌胃,祛瘀化浊汤组给予祛瘀化浊汤灌胃,正常组和模型组给予生理盐水灌胃,均3次/d,连续56 d。灌胃结束后取血检测胰岛素抵抗指数(HOMA-IR)、血脂(TC、TG、LDL-C、HDL-C)、肝功能(ALT、AST)和肝脏组织匀浆中TG、TC水平。结果模型组大鼠血清ALT、AST、TG、TC、LDL-C水平及HOMA-IR均明显高于正常组(P均<0.05),HDL-C水平明显低于正常组(P<0.05);多烯磷脂酰组和祛瘀化浊汤组血清ALT、AST、TG、TC、LDL-C水平及HOMA-IR均明显低于模型组(P均<0.05),HDL-C水平明显高于模型组(P均<0.05),且祛瘀化浊汤组各指标改善情况均明显优于多烯磷脂酰组(P均<0.05)。模型组大鼠肝组织匀浆中TC、TG水平显著高于正常组(P均<0.05),多烯磷脂酰组和祛瘀化浊汤组肝组织匀浆中TC、TG水平显著低于模型组(P均<0.05),且祛瘀化浊汤组明显低于多烯磷脂酰组(P均<0.05)。结论祛瘀化浊方能够改善NAFLD大鼠脂肪代谢紊乱,减轻肝脏脂质蓄积,纠正胰岛素抵抗。 Objective It is to observe the effect of decoction for resolving stasis and turbidity on lipid metabolism and liver function of rat models with non-alcoholic fatty liver (NAFLD) , and to explore its mechanism of the treatment for NAFLD. Methods 80 adult male SD rats were randomly divided into normal group, model group, polyene phosphatidyl group, and decoction for resolving stasis and turbidity group (treatment group) , each group had 20 rats. All the rats except in the normal group were fed with high fat fodder for 84d to establish NAFDL models. After the success of model establis- hing, polyene phosphatidyl group and treatment group were treated respectively with polyene phosphatidyl and decoction for resolving stasis and turbidity by gavage for 56 days, the normal group and model group were treated with normal saline by lavage for 56 days, all 3 times per day. After treatment, the levels of insulin resistance index (HOMA-IR) , blood lipid (TC, TG, LDL-C, HDL-C) , liver function (ALT, AST) in serum, and TG and TC in liver tissue homogenation were de- tected. Results The serum levels of ALT, AST, TG, TC, LDL-C and HOMA-IR were significantly higher while that of HDL-C was lower in the model group than those in the normal group (P 〈0. 05). The serum levels of ALT, AST, TG, TC, LDL-C and HOMA-IR were significantly lower while that of HDL-C was higher in polyene phosphatidyl group and treatment group than those in the model group (P 〈 0.05) , and the improvements of these indexes in the treatment group were better than that in polyene phosphatidyl group. The levels of TC and TG in liver tissue homogenate in the model group were significantly higher than those in the other three groups( P 〈 0.05 ) , and the levels of TC and TG in liver tissue homogenate in the treatment group were significantly lower than that in polyene phosphatidyl group( P 〈 0.05 ). Conclusion The decoction for resolving stasis and turbidity can improve fat metabolism disorder, reduce lipid accumulation in liver and correct insulin resistance in the rats with NAFLD.
