摘要
目的:探讨芦荟大黄素对肺癌A549细胞生长、迁移及F-actin的影响。方法:将肺癌A549细胞分为对照组、芦荟大黄素低剂量组(10μmol/L)、中剂量组(30μmol/L)和高剂量组(50μmol/L)。采用CCK-8法检测细胞增殖抑制率,Transwell小室检测细胞迁移能力,高内涵细胞成像系统分析芦荟大黄素对肺癌细胞骨架蛋白F-actin表达的影响,western blotting检测各组细胞小窝蛋白-1(Caveolin-1)、磷酸化内皮型一氧化氮合酶(p-eNOS)、p65的表达。结果:与对照组相比,芦荟大黄素低剂量组细胞增殖抑制率、迁移能力、细胞骨架蛋白的面积未见明显变化(P>0.05),而中、高剂量组细胞增殖抑制率增加、迁移能力降低,细骨胞架蛋白面积缩小,差异均有统计学意义(均P<0.05)。与对照组相比,低剂量组细胞中Caveolin-1、peNOS、p65蛋白的相对表达量并无明显变化(P>0.05);中、高剂量组细胞Caveolin-1、p65表达增多,p-eNOS表达减少,差异有统计学意义(P<0.05)。结论:芦荟大黄素能够抑制肺癌A549细胞的生长和迁移,其机制可能与影响Caveolin-1/eNOS/p65信号通路,增加F-actin的解聚、减少微丝的形成有关。
Objective: To investigate the effect and mechanism of aloe-emodin on the growth,migration and Factin of lung cancer A549 cells. Methods: The A549 cells were divided into control group,low-dose group(A549 cells +10 μmol/L aloe-emodin co-culture for 30 min),medium-dose group(A549 cells + 30 μmol/L aloe-emodin co-culture for 30 min)and high-dose group(A549 cells + 50 μmol/L aloe-emodin co-culture for 30 min). The cell proliferation rate was detected by CCK-8 assay. The migration ability of cells was detected by Transwell assay. The expressions of Caveolin-1,phosphorylated endothelial nitric oxide synthase(p-eNOS)and p65 were determined by western blotting. Results: The proliferation rate and migration ability of cells were markedly decreased in the medium-dose group and the high-dose group(P〈0. 05). The expression of Caveolin-1 and p65 was higher in medium and high dose groups than that in the control group,while the expression of peNOS was lower(P〈0. 05). However,no significant differences were found between the low-dose group and the control group(P〉0. 05). Conclusion: Aloe-emodin could inhibit the growth and migration of lung cancer A549 cells,and its mechanisms were by means of influencing Caveolin-1/eNOS/p65 signaling pathway and increasing F-actin depolymerization.
