摘要
目的测定贮藏过程中广金钱草种皮色素及相关酶活性变化,探讨种子贮藏过程中发生的生理生化变化,为揭示种子劣变机理,控制和评价种子质量提供依据。方法用紫外分光光度法测定叶绿素、类胡萝卜素、黑色素、花色素及络氨酸酶、多酚氧化酶、苯丙氨酸解氨酶活性。结果广金钱草种子在贮藏期间,叶绿素和类黄酮类色素含量随贮藏时间的延长而降低,类胡萝卜素含量变化不显著,黑色素含量随时间的延长而增加;从种皮颜色来看,叶绿素含量表现为绿色>黄色>黄褐色>深褐色>褐色,类胡萝卜素和类黄酮类色素表现为黄色>黄褐色>褐色>深褐色>绿色,黑色素表现为深褐色>褐色>黄褐色>黄色>绿色。络氨酸酶和多酚氧化酶活性随着贮藏时间的延长呈现上升趋势,苯丙氨酸解氨酶活性呈现降低趋势。从颜色来看,深褐色种皮络氨酸酶、多酚氧化酶分别是黄褐色种皮的1.43,2.47倍,黄褐色种皮苯丙氨酸解氨酶活性是褐色种皮的1.11倍。结论广金钱草在贮藏过程中,种皮络氨酸酶、多酚氧化酶、苯丙氨酸解氨酶活性的变化导致种皮色素含量的改变,从而引起种皮颜色的变化,最终导致种子发生劣变。
Objective To provide scientific basis for revealling the seed deterioration mechanism, controlling and evaluating the seed quality , the variation of pigment content and enzymes activities in seed coats of Desmodium styracifolium ( Osbeck. ) Merr. were studied during the storage process. Methods The content of chlorophyll, carotenoid, melanin and anthocyanin, and the ac- tivities of tyrosine Kinase, polyphenol oxidase and phenylalanine ammonia lyase were determined by ultraviolet spectrophotometry. Results With the extension of storage time, the content of chlorophyll and fiavonoid decreased, the change of carotenoid content is not obvious, the content of melanin increased, Compared with chlorophyll content, green seed 〉 yellow seed 〉 yellowish - brown seed 〉 dark brown seed 〉 brown seed. Compared with the content of carotenoid and flavonoid pigments , yellow seed 〉 yellowish - brown seed 〉 brown seed 〉 dark brown seed 〉 green seed. Compared with Melanin content, dark brown seed 〉 brown seed 〉 yellow seed 〉 green seed. The activities of tyrosine kinase and polyphenol oxidase asended and the activity of phenylalanine ammo- nialyase desended with the elongation of storage time, the content of tyrosinase and polyphenol oxidase in clark brown seed coat were 1.43 and 2.47 times of that of yellowish - brown seeds coat, the content of phenylalanine ammonia lyase in yellowish - brown seed coat was 1.11 times of that of brown seed coat. Conclusion In the process of storage, the activities variation of the tyrosine kinase, polyphenol oxidase and phenylalanine ammonialyase resulted in the content variation of pigment in seed coat, which caused the changes in color of seed coat and finally led to the seed deterioration.
