摘要
目的探讨蒜氨酸在家兔体内的代谢、分布与排泄情况。方法通过静脉注射蒜氨酸后,不同时间采集家兔血液,采用高效液相色谱法检测蒜氨酸和代谢产物大蒜素;并在血药浓度较高时,处死家兔,收集内脏器官,检测蒜氨酸和大蒜素。结果蒜氨酸血药浓度-时间变化曲线的曲线下面积(AUC)为16 964.0(mg·min)/L,半衰期[t_(1/2(12))]为237.3 min、t_(1/2(21))为192.5 min,表观分布容积(Vd)为4.912 L,清除率(Cl)为0.017 7 mg/(L?min)。大蒜素AUC为12 583.1(mg·min)/L,t_(1/2(12))为227.2 min,t_(1/2(21))为260.53 min,Vd为4.51 L,Cl为0.012 mg/(L?min)。结论蒜氨酸、大蒜素药代动力学均符合二房室一级动力学过程,体液分布以大蒜素通过呼吸道排泄。
Objective To study the alliin in rabbit in vivo metabolism,distribution and excretion. Methods After intravenous injection of alliin,collectted rabbit blood at different time,used high performance liquid chromatography(HPLC)detection of alliin and allicin and metabolites;and putted to death on rabbit when the blood drug concentration high,collectted the internal organs,detected of alliin and allicin. Results The area under the curve(AUC)of the allicine blood concentrationtime curve was 16 964.0(mg·min)/L,half-life(t_(1/2(12))was 237.33 min,t_(1/2(21))was 192.5 min,the distribution of apparent volume (Vd)was 4.912 L,clearance(Cl)was 0.017 7 mg(L?min). Allicin AUC was 12 583.1(mg·min)/L,t_(1/2(12))was 227.2 min,t_(1/2(21))was 260.53 min,Vd was 4.51 L,Cl was 0.012 mg/(L?min).Conclusion Alliin,allicin pharmacokinetic conforms to the second room firt order kinetics process,the distribution of body fluids is excreted by allicin through the respiratory tract. The study to provide theoretical basis for study of the development of alliin.
