摘要
目的:研究益脾养肝方对二乙基亚硝胺(DEN)诱导的大鼠肝癌癌前病变病理组织和NF-κB、STAT3蛋白表达的影响。方法:将大鼠随机分成对照组、模型组、益脾养肝方大、小剂量组。采用腹腔注射DEN诱发大鼠肝癌前病变模型,给药16周后处死。观察大鼠一般情况和体质量,肝功变化,肝组织HE染色以及免疫组化检测NF-κB和STAT3蛋白表达。结果:与模型组比较,YPYGF大、小剂量组的ALT、AST和GGT均明显降低,其中YPYGF大剂量组效果最佳(P〈0.01)。病理组织结果显示,益脾养肝方可以改善大鼠肝癌癌前病变的病理形态,能显著抑制肝组织NF-κB和STAT3的蛋白表达。结论:益脾养肝方对于大鼠肝癌癌前病变具有抑制作用,其作用可能与抑制炎症因子NF-κB和STAT3表达有关。
Objective: To study the effect of YPYGF on the amelioration of pathological morphology and expression of NF-κB and STAT3 proteins in diethylnitrosamine(DEN) induced rat liver precancerous lesion. Methods: The hepatic precancerous lesion model of male Wistar rats induced by intraperitoneal injection of DEN was used. Rats were randomly divided into the normal control,the DEN group,and two YPYGF(0. 5 g/m L and 1. 0 g/m L) treatment groups. After 16 weeks of administration,rats were sacrificed. We observed generral conditions,body weight and liver function of rats. Liver tissues were measured by HE staining,and the expressions of NF-κB and STAT3 proteins of liver tissues were measured by immunohistochemical detection. Results:Compared with model group,ALT,AST and GGT of two YPYGF groups were significantly lower and the best was high-dose group of YPYGF(P 0. 01). YPYGF can improve the rat liver precancerous pathological morphology,and can remarkably inhibit the expressions of liver tissue NF-κB and STAT3 proteins. Conclusion: The results suggest that YPYGF significantly suppress precancerous lesion of rats' liver by mediating expressions of NF-κB and STAT3 proteins.
