摘要
目的:研究甘草酸对UVB光老化人皮肤角质形成细胞(Ha Ca T)的保护作用及炎症因子影响。方法:采用30 m J/cm2的UVB照射Ha Ca T细胞,建立光老化模型;用10-7、10-6、10-5mol/L甘草酸处理光老化细胞24 h。MTT法检测细胞增殖率;采用试剂盒检测各组细胞SOD、GSH及CAT活性;Western Blot法检测各组细胞IL-1β、IL-6、TNF-α蛋白表达量。结果:与空白组相比,模型组GSH、SOD及CAT活性显著降低(P<0.01),IL-1β、IL-6、TNF-α蛋白表达量显著升高(P<0.01);与模型组相比,不同浓度甘草酸可使SOD、GSH及CAT活性显著升高(P<0.01),IL-1β、IL-6、TNF-α蛋白表达量显著降低(P<0.05,P<0.01)。结论:甘草酸能保护UVB诱导的Ha Ca T细胞光老化,其机制可能与其抑制氧化损伤,减少炎症因子的产生有关。
Objective To study the protective role of glycyrrhizic acid on light aging cells of HaCaT and its in-fluence to inflammatory cytokines. Methods; Light aging model was set up by the irradiation of ultraviolet B (UVB ) at irradiation of 30 mJ/cm2 ; light aging cells were intervened witli diferent concerhizic acid(10 7 mol/L ^,10 6 mol/L^,and 10 5 mol/L ^) for 24 h. Cell proliferation rates were MTT method,the levels of SOD, GSH,and CAT were tested in each group, the expressions of IL - 1 , IL - 6 ,and TNF - a were examined by Western blot method. Results; Compared to the SOD,GSH,and CAT were significantly decreased(〉 〈0.01); the expression levels of IL - 1 #,IL - 6 , andTNF - a were greatly increased(〉 〈 0. 01) . Compared / the model group, the activitCAT were significantly raised( P 〈 0. 01),the levels of IL , - 1 # , IL - 6 and TNF - a were obviously decreased (P 〈0 .05,〉 〈 0. 01) witii diferent concentration of glycyrrhizic acid. Conclusion ; Glycyrrhizic acid can sig-nificantly protect light aging cells of HaCaT induced by UVB,which mechanism may be related to suppressing the oxidative damage and inhibiting the secretion of inflammatory cytokines.
