摘要
目的改良大鼠肺微血管内皮细胞（PMVECs）原代培养方法，并对所培养的PMVECs进行鉴定。方法组织贴块法培养PMVECs，从动物的选取、肺灌注、细胞漂洗次数及试剂温度等方面进行了改良。免疫组织化学鉴定PMVECs。结果原代培养的PMVECs呈典型的铺路石样生长。免疫组织化学显示Ⅷ因子及血管性血友病因子表达阳性。结论经改良后培养PMVECs，细胞培养的重复性高且生长状态好。
Objective To improve the method of primary cultivation of rat pulmonary microvascular endothelial cells （PMVECs）, and identify the primary cultivated PMVECs. Methods Tissue block adhering wall method was used to cultivate PMVECs. Primary cultivation method was improved from the procedures of animal selection, pulmonary perfusion, the number of cells rinses and temperature of reagents and so on. Immunohistochemical obserbation PMVECs. Results The PMVECs were exhibited as polygon. Immunohistochemical staining revealed that expression of Ⅷ and von willebrand factor was positive. Conclusions After the improved method of PMVECs, the reproducibility of cell culture is high and the growth status is good.
