摘要
目的检测CD_(11c)^+DC(DC_1)和CD_(123)^+DC(DC_2)在鼻息肉组织中的表达,探讨树突状细胞亚群在鼻息肉发病机理中的意义。方法用HE染色法观察标本炎性细胞浸润情况。免疫组化的方法检测鼻息肉组和下鼻甲组标本中CD_(11c)^+DC(DC_1)和CD_(123)^+DC(DC_2)的表达情况。结果 70%的鼻息肉以淋巴细胞和浆细胞浸润为主,30%的鼻息肉以嗜酸性粒细胞浸润为主。CD_(11c)和CD_(123)在鼻息肉组的表达均高于下鼻甲组,差别有高度统计意义(P<0.01)。在鼻息肉组织中,CD_(11c)和CD_(123)的表达差别无显著性,CD_(11c)^+DC与CD_(123)^+DC数量的比较,差别有统计意义(P<0.05)。结论潮汕地区鼻息肉70%以淋巴细胞和浆细胞浸润为主。鼻息肉组织中存在DC1/DC2混合模式。DC1在鼻息肉中占优势地位,诱导Th1型免疫反应为主导免疫,最终导致鼻息肉中Th_1/Th_2免疫应答失衡。
Objective To explore the role of DC1 and DC2 in the pathogenesis of human nasal polyps. Methods The specimens of nasal polyp and nasal septum were studied by hematoxylin and eosin staining (HE). The expression of CD11C+DC and CD123+DC in all the samples were detected by two-step immunohistochemistry. Results Seventy percent of the samples consisted of abundant T lymphocytes and plasma cells and thirty percent of samples were dominated by eosinophils. Both of the expression of CD11c+DC and CD123+DC in nasal polyp tissues were significantly higher than those of control group (P〈0.01). In nasal polyps, there are no significant difference between the expression of CD11c+DC and CD123+DC, but the quantity of CD11c+DC was significantly larger than that of CD123+DC (P〈0.05). Conclusions Seventy percent of the nasal polyps in Chaoshan area are dominatly infiltrating lymphocytes and plasma cells. There is a mixed DC1/DC2 pattern in nasal polyps. DC1 cells are in the dominant position in nasal polyps and induce the Th1 type immune response, which lead to Th1/Th2immunity imbalance.
