bel-2结合抗凋亡基因1及表皮生长因子受体在三阴性乳腺癌中的表达 被引量:4

Expressions of bcl-2-associated athanogene 1 and epithelial growth factor receptor gene in triple negative breast cancer

摘要 目的检测bcl-2结合抗凋亡基因1(BAG-1)和表皮生长因子受体(EGFR)mRNA在三阴性乳腺癌(TNBC)中的表达情况。方法收集十堰市太和医院2013年10月至2014年8月收治的TNBC患者51例,采用实时定量聚合酶链反应检测TNBC组织及其部分对应癌旁组织中BAG-1 mRNA和EGFR mRNA的表达,分析基因表达与TNBC患者临床病理参数之间的关系。结果BAG-1 mRNA和EGFR mRNA在TNBC组织中均为高表达状态,相对表达量分别为0.57±0.25、0.61±0.21,其在TNBC组织中的表达水平均高于癌旁组织(0.19±0.12、0.21±0.11),差异均有统计学意义(t值分别为5.12、7.17,均P〈0.001)。不同临床分期、淋巴结转移TNBC患者的BAG-1 mRNA和EGFR mRNA表达水平比较,差异均有统计学意义(均P〈0.05),而不同肿瘤直径、年龄的患者两种基因mRNA表达水平差异均无统计学意义(均P〉0.05)。结论BAG-1 mRNA、EGFR mRNA在TNBC中呈高表达状态,其高表达可能与患者不良预后相关,可作为预测TNBC患者预后的分子指标。 ObjectiveTo investigate mRNA expressions of bcl-2-associated athanogene1 (BAG-1), epithelial growth factor receptor (EGFR) in triple negative breast cancer (TNBC).MethodsPolymerase chain reaction (PCR) method was used to detect mRNA expression levels of BAG-1 and EGFR in 51 TNBC cases combined with adjacent tissues in Taihe Hospital of Shiyan City from October 2013 to August 2014, and the relationship between gene expression and the clinicopathologic parameters of TNBC patients was analyzed.ResultsThe relative expressions of BAG-1 mRNA and EGFR mRNA were 0.57±0.25, 0.61±0.21 respectively in TNBC. The expression level of mRNA in breast cancer (0.19±0.12, t = 5.12) was higher than that in adjacent tissues (0.21±0.11, t = 7.17), and there was no significant difference (P 〈 0.001). The difference of expressions of BAG-1 mRNA and EGFR mRNA in TNBC patients with different clinical stage or lymph node metastasis were statistically significant (all P 〈 0.05), but there was no significant difference in mRNA expression level of the patients with different tumor diameter or age (all P 〉 0.05).ConclusionBAG-1 mRNA, EGFR mRNA are highly expressed in TNBC, which may be related with the poor prognosis and may be a molecular index for predicting the prognosis of TNBC patients.
