摘要
目的探讨花椒毒酚对重症急性胰腺炎(SAP)大鼠脑损伤的保护作用及对核因子-κB/诱导型一氧化氮合酶(NF-κB/iNOS)通路的影响。方法将64只Wistar大鼠随机分为8组:正常对照3 h组、12 h组,模型3 h组、12 h组,花椒毒酚低剂量(5 mg/kg)3 h组、12 h组,花椒毒酚高剂量(10 mg/kg)3 h组、12 h组,每组8只;采用5%牛黄胆酸钠(2 mL/kg)注入胰胆管进行SAP造模,正常对照组为假手术组。于术后3、12 h收集各组大鼠腹主动脉血、胰腺组织和脑组织,观察各组大鼠胰腺组织和脑组织HE染色评分;检测各组大鼠血浆淀粉酶、NO、NF-κB p65水平及脑组织含水量、神经功能评分、NF-κB p65、iNOS蛋白相对表达量。结果模型组大鼠术后3、12 h胰腺组织HE染色评分、血浆淀粉酶均较对照组明显升高(P<0.05),花椒毒酚高、低剂量组胰腺组织HE染色评分、血浆淀粉酶均较模型组明显降低(P<0.05),且高剂量组各指标均明显低于低剂量组(P<0.05);模型组大鼠术后3、12 h脑组织HE染色评分、脑组织含水量、神经功能评分均较对照组明显升高(P<0.05),花椒毒酚高低剂量组胰腺组织、脑组织HE染色评分、脑组织含水量、神经功能评分均较模型组明显降低(P<0.05),且高剂量组各指标均明显低于低剂量组(P<0.05);模型组大鼠术后3、12 h血清NO、NF-κB p65水平及脑组织NF-κB p65、iNOS水平均明显高于对照组(P<0.05),花椒毒酚高、低剂量组各指标均较模型组明显降低(P<0.05),且高剂量组各指标下降幅度尤为明显(P<0.05)。结论花椒毒酚能够通过降低SAP大鼠脑组织NF-κB、iNOS蛋白表达,发挥对大鼠脑损伤的保护作用。
Objective To explore the protective effect of zanthoxylum on brain injury and its influence on NF-κB/iNOS pathway in rats with severe acute pancreatitis(SAP).Methods Totally 64 Wistar rats were randomly divided into 8 groups,the normal control 3 h group,12 h group,the model 3 h group,12 h group,low-dose paracetamol(5 mg/kg) 3 h group,12 h group,high-dose paracetamol(10 mg/kg) 3 h group,12 h group,8 rats in each group.The5% sodium taurocholate(2 mL/kg) was injected into pancreaticobiliary duct to build SAP model,and the normal control group was as sham operation group.Abdominal aorta blood,pancreatic tissue and brain tissue were collected at postoperative 3 h and 12 h in each group.HE staining score of pancreatic and brain tissue of rats was observed in each group,and plasma amylase,NO,NF-κB p65 levels,brain tissue water content,neurological function score,and relative expression of NF-κB p65 and iNOS proteins were detected in rats of each group.Results HE staining score of pancreatic tissue and plasma amylase at postoperative 3 h and 12 h in the model group were significantly higher than those of control group(P 〈 0.05),and those in paracetamol low-dose and high-dose groups were significantly lower than those of model group(P 〈 0.05),and those of high-dose group were significantly lower than those of low-dose group(P 〈0.05).HE staining score of brain tissue,water content of brain tissue and neurological function scores at postoperative3 h and 12 h in the model group were significantly higher than those of control group(P 〈 0.05),and those in paracetamol low-dose and high-dose groups were significantly lower than those of model group(P 〈 0.05),and those of high-dose group were significantly lower than those of low-dose group(P 〈 0.05).Serum NO,NF-κB p65,and brain tissue NF-κB p65,iNOS levels were significantly higher than those of control group(P 〈 0.05),and those in paracetamol low-dose and high-dose groups were significantly lower than those of model group(P 〈 0.05),and the decline in high-dose group were more obvious(P 〈 0.05).Conclusion Zanthoxylum can protect the brain of rats from injury by reducing the expression of NF-κB and iNOS protein in brain tissue of SAP rats.
