摘要
采用超高效液相色谱建立鸡肉中色氨酸含量检测方法。鸡肉样品经过5 mol/L氢氧化锂碱解提取,经Acquity UPLC BEH C18(50 mm×2.1 mm,1.7μm)分离,以甲醇和0.0085 mol/L乙酸钠缓冲溶液(pH4.0)为流动相进行梯度洗脱,PDA检测器,280 nm波长采集数据,在(0.2~100.0)mg/L浓度范围内,线性良好,相关系数0.99996,方法的最低定量限为20 mg/kg,通过1、2、4 mg/g3个浓度的加标回收实验表明,回收率为87.8%~98.8%,RSD值为0.97%~2.13%。该方法操作简单、准确可靠,适用于鸡肉中色氨酸的定量测定。
A method for the determination of tryptophan in chicken was established by Ultra performance liquid chromatography. In this method, chicken samples were extracted with 5 mol/L lithium hydroxide solution. Tryptophan was analyzed by Acquity UPLC BEH C18 column(50 mm×2.1 mm, 1.7 μm) using methanol and 0.0085 mol/L sodium acetate buffer solution(pH4.0) as the mobile phases. The analyte was detected with PDA detector at 280 nm as detection wavelength. The calibration curve of tryptophan was linear in a concentration range of(0.2-100.0)mg/L(r=0.99996), and the limits of quantitation was 20 mg/kg. The recoveries of 1, 2, 4 mg/g fortified samples ranged from 87.8% to 98.8%, with RSDs of 0.97% to 2.13%. The established methods were proved accurate and reliable which can meet the requirements of the detection of tryptophan in chicken.
