扶正祛浊法对糖尿病肾病大鼠TLR4/MyD88通路的影响 被引量:2

Effect of Chinese Medical Method of Fuzheng Quzhuo on the Pathyway TLR4/MyD88 in Rats with Diabetic Nephropathy

摘要 目的观察研究扶正祛浊法治疗糖尿病肾病(DN)模型大鼠的疗效和作用机制。方法 SD大鼠60只,随机分为空白组、DN组、洛汀新组[2mg/(kg·d)]、扶正祛浊方低(1.32g/mL)、中(2.64g/mL)、高(5.28g/mL)剂量组,各10只,分别予生理盐水及相应药物灌胃,造模前后以及给药后4周、8周检测各组大鼠血糖、肾功能、尿蛋白等;8周后Western blot法检测大鼠肾组织中Toll样受体4(TLR4)、髓样分化因子88(MyD88)蛋白水平。结果与DN组比较,扶正祛浊方低、中、高剂量组及洛汀新组大鼠血肌酐水平、24h尿微量蛋白量及肾组织My D88蛋白表达水平明显下降[(32.50±2.95)umol/L、(29.17±3.87)umol/L、(29.00±4.65)umol/L、(35.50±5.47)umol/L比(53.67±3.88)umol/L,P<0.05;(53.25±6.84)mg/d、(47.69±6.67)mg/d、(33.52±4.75)mg/d、(51.08±3.82)mg/d比(62.89±3.69)mg/d,P<0.05;(0.1703±0.0195)、(0.1592±0.0178)、(0.1349±0.0121)、(0.1221±0.0162)比(0.2055±0.0194),P<0.05];扶正祛浊方高剂量组、洛汀新组大鼠尿素氮水平均有明显下降[(7.43±1.09)mmol/L、(10.07±0.85)mmol/L比(11.02±0.87)mmol/L,P<0.05];扶正祛浊方中、高剂量组,洛汀新组大鼠TLR4蛋白表达水平有明显下降[(0.1613±0.0338)、(0.1336±0.0165)、(0.1156±0.0072)比(0.2227±0.0449),P<0.05]。结论扶正祛浊法对糖尿病肾脏炎症反应有抑制作用,从而对DN大鼠肾脏产生保护作用。 Objective To study the effect of Fuzheng Quzhuo(Strengthening Zheng and Eliminating Turbid)method on diabetic nephropathy in rats and its mechanism. Methods Sixty SD rats were randomized into blank group, DN model group, benazepril hydrochloride tablet group(2 mg/(kg·d)), low-, medium-, and high-dose of Fuzheng Quzhuo Decoction groups(1.32, 2.64, 5.28 g/mL), with 10 rats in each group. The drug groups were given the corresponding drug and blank and DN model groups was given saline by gavage. Blood sugar, renal function,and urinary protein were detected before and after modeling and before treatment and at 4, 8 weeks after treatment. At the end of the experiment(at 8 weeks) Western Blot was used to determine the level of Toll-like receptor 4(TLR4) and myeloid differentiation factor 88(MyD88) in renal tissues of the rat. Results Compared with DN model group, the low-, medium-, and high-dose of Fuzheng Quzhuo Decoction groups and benazepril hydrochloride tablet group had obvious decrease in the creatine level(32.50±2.95μmol/L, 29.17±3.87μmol/L, 29.00±4.65μmol/L, 35.50 ±5.47μmol/L vs 53.67 ±3.88μmol/L, P 0.05), 24 h microalbuminuria(53.25 ±6.84 mg/d, 47.69 ±6.67 mg/d, 33.52±4.75 mg/d, 51.08±3.83 mg/d vs 62.89±3.69 mg/d, P0.05), and the level of MyD88 protein in renal tissue(0.1703±0.0195, 0.1592±0.0178, 0.1349±0.0121, 0.1221±0.0162 vs 0.2055±0.0194, P0.05; the high-dose of Fuzheng Quzhuo Decoction group and benazepril hydrochloride tablet group had reduction in urea nitrogen(7.43±1.09 mmol/L, 10.07 ±0.85 mmol/L vs 11.02 ±0.87 mmol/L, P 0.05); the medium-and high-dose of Fuzheng Quzhuo Decoction groups and benazepril hydrochloride tablet group had downregulation of TLR4 protein(0.1613 ±0.0338,0.1336 ±0.0165, 0.1156 ±0.0072 vs 0.2227 ±0.0449, P0.05). Conclusion Fuzheng Quzhuo method can protect the renal function in diabetic nephropathy rats by suppressing the renal inflammation.
