摘要
目的通过分析五倍子等中药及其组方对女性生殖道感染厌氧菌耐药菌株的抑制作用,筛选抑菌活性强的中药。方法应用平皿打孔法测定五倍子等中药及其组方对厌氧菌耐药菌株的抑菌环直径;应用微量肉汤稀释法测定五味子、五倍子、黄连、大黄不同组方对脆弱类杆菌耐药菌株的最低抑菌浓度(MIC),并且分析五味子等4个组方对耐药菌株是否有协同抑菌作用。结果五倍子、诃子分别对厌氧消化链球菌、脆弱类杆菌耐药菌株的抑菌环直径最大,分别为21.33 mm和20.38 mm。五倍子、诃子对消化链球菌耐药菌株的抑菌环直径明显高于黄连、金银花、大黄(P<0.01);诃子、黄连与大黄、金银花,五倍子、诃子、黄连、大黄与金银花对脆弱类杆菌耐药菌株的抑菌环直径差异有统计学意义(P<0.05,P<0.01)。五倍子与黄连、大黄组方,诃子与黄连、大黄组方对消化链球菌耐药菌株的抑菌环直径均值均明显低于其单味药的均值(P<0.01);五倍子与大黄,黄连与诃子,诃子与黄连、大黄组方对脆弱类杆菌耐药菌株的抑菌环直径均值明显低于其单味药均值(P<0.05,P<0.01)。5种中药对消化链球菌和脆弱类杆菌耐药菌株的敏感率最高者分别是五倍子(93.33%)和诃子(70.83%)。五味子与大黄、黄连、五倍子组方对脆弱类杆菌耐药菌株的抑制浓度指数(FIC)分别为0.28、0.31、0.75。五倍子与大黄组方的FIC值为1.13。结论五倍子、诃子对消化链球菌、脆弱类杆菌耐药菌株具有较强的抗菌活性。五味子与大黄、黄连、五倍子组方对脆弱类杆菌耐药菌株具有协同抑菌作用。有必要对中药及其组方对厌氧菌耐药菌的作用及其机制作进一步的深入研究。
Objective To screen the traditional Chinese medicine with strong antibacterial activity by analyzing the inhibition effect of Chinese gall and the Chinese herbal formula on anaerobic drug-resistant strains in women with reproductive tract infection. Methods The inhibition zone diameter of anaerobic drug-resistant strains was detected by plate-perforation method. The minimal inhibitory concentrations of Schisandra chinensis,Chinese gall,Coptis chinensis,and Rheum officinale to Bacteriodes fragilis drug-resistant strains were determined by microdilution broth method,the synergistic antibacterial effects of four Chinese herbal formulas for drug-resistant strains were analyzed.Results The inhibition zone diameters of Chinese Gall and myrobalan to anaerobic peptostreptococcus and Bacteriodes fragilis drug-resistant strains were the largest,respectively,which were 21. 33 mm and 20. 38 mm,respectively. The inhibition zone diameters of Chinese Gall and myrobalan to peptostreptococcus were statistically significantly higher than those of Coptis chinensis,honeysuckle,and rheum officinale(P〈0. 01); there was statistically significant difference in the inhibition zone diameter among myrobalan,Coptis chinensis and rheum officinale,honeysuckle,Chinese Gall,myrobalan,Coptis chinensis,rheum officinale and honeysuckle to Bacteriodes fragilis drug-resistant strains(P〈0. 05,P〈0. 01). The average inhibition zone diameters of Chinese gall and Coptis chinensis,rheum officinale formula,fructus chebulae,Coptis chinensis,and rheum officinale formula to peptostreptococcus drug-resistant strains were statistically significantly lower than those to single traditional Chinese medicine(P〈0. 01). The average inhibition zone diameters of Chinese gall and rheum officinale,Coptis chinensis and fructus chebulae,fructus chebulae and Coptis chinensis,rheum officinale formulas to Bacteriodes fragilis drug-resistant strains were statistically significantly lower than those to single traditional Chinese medicine(P〈0. 05,P〈0. 01). The sensitivies of Chinese gall and fructus chebulae to peptostreptococcus and Bacteriodes fragilis drug-resistant strains were the highest,which were 93. 33% and 70. 83%,respectively. The fractional inhibitory concentration indexes of Schisandra chinensis and rheum officinale,Coptis chinensis,Chinese gall formulas to Bacteriodes fragilis drug-resistant strains were 0. 28,0. 31,and 0. 75,respectively. Fractional inhibitory concentration index of Chinese gall and rheum officinale formula was 1. 13. Conclusion Chinese gall and fructus chebulae have strong antibacterial activity to peptostreptococcus and Bacteriodes fragilis drug-resistant strains. Schisandra chinensis and rheum officinale,Coptis chinensis,Chinese gall formula has synergistic antibacterial effect. It is necessary to further research the inhibitory effect and mechanism of traditional Chinese medicine and Chinese herbal formula to anaerobic drug-resistant strains.
