摘要
目的建立高效液相色谱法(high performance liquid chromatography,HPLC)测定葡萄酒中柠檬酸的含量的分析方法。方法以0.02 mol/L甲醇和磷酸二氢钾(5:95,V:V)为流动相,流速为0.7 m L/min,在此色谱条件下,采用二级管阵列检测器进行检测。结果在0.05~1.00 g/L范围内,浓度与峰面积的线性关系良好,相关系数为0.9999。在加标浓度为5.0、20.0、50.0 g/L时的回收率为93.9%~106.3%,相对标准偏差(relative standard deviation,RSD)为1.67%~2.99%之间。结论该方法具有较高的灵敏度和准确性,且操作简单,适用于检测葡萄酒样品中的柠檬酸。
Objective To establish a method for determination of the content of citric acid in wine by high performance liquid chromatography method(HPLC). Methods The mobile phase was 0.02 mol/L methanol and potassium dihydrogen phosphate(5:95, V:V), and the flow rate was 0.7 m L/min. The sample was detected by the secondary tube array detector. Results In the range of 0.05-1.00 g/L, the linear relationship between the concentration of citric acid and the peak area was good, and the correlation coefficient was 0.9999. At the spiked levels of 5, 20, and 50 g/L, the recovery rates were 93.9%-106.3%, and the relative standard deviation(RSD) was 1.67%-2.99%. Conclusion This method has high sensitivity and accuracy, and simple operation, which is suitable for detecting citric acid in wine samples.
