摘要
对淡紫拟青霉TD16色素结构和性质进行研究。将淡紫拟青霉TD16菌株进行液态发酵,发酵液经乙酸乙酯萃取、蒸干、甲醇复溶、过滤除杂获得色素样品。对所得样品进行定性分析、紫外-可见光谱分析、红外光谱分析和液质联用分析,并测定其稳定性和抗氧化性。结果表明,TD16色素是一种相对分子质量为396.01的羟基蒽醌类化合物,该色素在高温、中等光强和酸性条件下稳定,对大多数金属离子不敏感,但Fe^(3+)和Cu^(2+)会造成其变色和褪色,Fe^(2+)略有增色作用,对羟基自由基和超氧阴离子均有较强清除能力。
The structure, stability and antioxidant activity of the pigment secreted from Paecilomyces lilacinus TD 16 was studied. The pigment was separated by ethyl acetate/water liquid-liquid extraction, and purified by re-dissolution with methanol and filtration. Based on the qualitative analysis, UV-vis, FFIR and LC-MS analysis, the structure of TD16 pigment was preliminarily characterized as hydroxyanthraquinone compound with the relative molecular mass of 396.01. TD16 pigment was stable on high temperature, medium irradiance and low pH, and insensitive to most of the metal ions. The color of TD16 pigment changed in the presence of Cu2+ and Fe3+ions but was enhanced by Fe2+. Moreover, TD16 pigment exhibited a high antioxidant activity of scavenging hydroxyl radical and superoxide radical.
