快速溶剂萃取-高效液相色谱法联用测定坚果中α维生素E的含量 被引量:2

Determination ofα-Vitamin E in nuts by high performance liquid chromatography with accelerated solvent extraction

摘要 利用快速溶剂萃取与高效液相色谱技术联用技术,建立了坚果中α维生素E(α-VE)含量的测定方法。以杏仁为例,考察了萃取α-VE的工艺条件,包括萃取溶剂、温度、静态萃取时间和循环次数等。结果表明最佳萃取工艺为:正己烷为萃取溶剂,温度110℃,萃取时间300s,循环2次。分离条件为:以甲醇-水溶液作为流动相进行梯度洗脱,C18色谱柱分离,检测波长:294nm。结果显示,在2~100μg/mL范围内,α-VE的峰面积与其质量浓度线性关系良好(r=0.9999),平均加标回收率为90.1~97.2%,检出限为0.04μg/mL。按照优化后的萃取方法测定了生花生,山核桃,杏仁以及葵花籽4种坚果样品,α-VE的含量为1.82~154.65μg/g。快速溶剂萃取法方法简单,使用溶剂量少,萃取时间短,回收率稳定,满足测定坚果中αVE的检测要求。 The accelerated solvent extraction parameters(extraction solvent,temperature,statictime and cycles)of almonds were optimized in order to maximize the extraction efficiency.The results indicated that the optimum conditions for extraction were as follows:extraction temperature of 110℃,static extraction time of 300 sand static loop of two times while using hexane as the solvent.For the HPLC determination,a C18 column was used,and a mixture of methanol-water was adopted as the mobile phase.The quantification was based on UV detection at 294 nm.The calibration curves ofα-VE was in good linearity over the range of 2-100μg/mL(r=0.9999),the average recoveries ofα-VE was 90.1%-97.2%and the limit of detection was 0.04μg/mL.Theα-VE contents of peanuts,pecans,almondsand,sunflower seed samples based on the optimized extraction method were 1.82-154.65μg/g.The accelerated solvent extraction is with the advantages of less solvent,less time and stable recoveries,is of celerity and simpleicity for the detection ofα-VE.
