摘要
目的:建立了HPLC-DAD法检测中成药及保健食品中16种降糖类化学药物的测定方法。方法:样品经甲醇提取,采用C18色谱柱(250 mm×4.6 mm,5μm),以甲醇-0.01 mol/L乙酸铵溶液为流动相,梯度洗脱,检测波长210nm、235 nm,外标法定量。结果:16种降糖类化学药物在0.005 mg·m L^-1-0.040 mg·m L^-1浓度范围内线性关系良好,回收率93.1%-108.4%,检出限为0.0003 mg·m L^-1-0.0027 mg·m L^-1。结论:本方法灵敏度高、选择性好、准确度高,可更好的为日常监督检验工作提供技术支持。
Objective: To establish an HPLC-DAD method for the simultaneous determination of sixteen anti-diabetic agents illegally added in traditional Chinese medicines and health food. Methods: Samples were extracted with methanol,and separated by Hplc and quantified with external standard method. A C18 column(250 mm × 4. 6 mm,5 μm) was used,with the mobile phase of methanol and 0. 01 mol/L ammonium acetate by gradient elution. The detection wavelengths were 210 nm and 235 nm. Results:The calibration curves for the sixteen synthetic anti-diabetic agents were linear in the concentration ranges of 0. 005 mg·m L^-1-0. 040 mg·m L^-1 and the recoveries were 93. 1%-108. 4% respectively,and the detection limits of sixteen anti-diabetic agents were 0. 0003 mg·m L-1-0. 0027 mg·m L^-1. Conclusion: The method is sensitive and selective,with excellent accuracy. It can be used for the routine detection in health food.
