摘要
以新疆野苹果成熟胚为试材,采用组织培养的方法,研究了新疆野苹果增殖及生根的最优培养基。结果表明:最适的增殖培养基为MS+0.4 mg·L^(-1) 6-BA+0.1mg·L^(-1) NAA,最适的生根培养基为1/2MS+1.0mg·L^(-1) NAA,组培苗利用水培练苗后移栽成活率为65%。该研究建立了以新疆野苹果成熟胚为外植体的快速繁殖体系,并比较了新疆野苹果在不同增殖、生根培养基中的生长情况,以期为新疆野苹果的研究和保护提供帮助,为其快速繁殖和进一步的遗传转化提供参考依据。
The seed embryo of Malus sieversii was used as experimental materials,through tissue culture method the optimum medium for proliferation and rooting of Malus sieversii were stuided.The results showed that the optimal proliferation medium was MS+0.4 mg·L^-1 6-BA+0.1 mg·L^-1 NAA.Rooting medium was 1/2 MS+1.0 mg L^-1 NAA.The survival rate of seedlings would raise to65% by hardening-seedling with hydroponic culture before transplant to soil.The research laid a foundation for obtaining plenty of health seedlings in short time,which greatly benefited for wild apple forest ecological reconstruction in Tianshan Mountains,since they were seriously destroyed in past10 years.
