ICP-MS法同时测定林下西洋参和园地栽培西洋参中14种微量元素的含量 被引量:6

Simultaneous Determination of 14 Kinds of Microelements in Underwood-cultivated and Garden-cultivated Panax quinquefolius by ICP-MS

摘要 目的:建立同时测定林下西洋参和园地栽培西洋参中14种微量元素含量的方法。方法:采用电感耦合等离子体质谱法。射频功率为1 350 W,采样深度为7.0 mm,载气流量为1.1 L/min,雾化室温度为2℃,蠕动泵转速分辨率为0.1 rps,等离子体气流量为15.0 L/min,雾化器为高盐雾化器,氧化物指标(氧化铈离子/铈)<0.5%,双电荷指标(二价钡离子/一价钡离子)<1%;微波消解功率为3 600 W,升温程序为190℃保持48 h,冷却至室温,再升温至150℃,保持12 h。结果:锰、铬、锶、锌、铷、硼、镍、铜、钒、锡、砷、钼、硒、钴元素检测质量浓度线性范围分别为0.1~500μg/L(r=0.999 8)、0.5~500μg/L(r=0.999 9)、0.1~500μg/L(r=0.999 9)、0.5~500μg/L(r=0.999 9)、0.5~500μg/L(r=0.999 9)、0.1~200μg/L(r=0.999 9)、0.5~500μg/L(r=0.999 9)、1~500μg/L(r=0.999 9)、0.1~500μg/L(r=0.999 8)、0.1~200μg/L(r=0.999 8)、0.5~500μg/L(r=0.999 9)、0.1~200μg/L(r=0.999 8)、0.5~500μg/L(r=0.999 9)、0.1~500μg/L(r=0.999 9);定量限均小于0.60μg/L,检测限均小于0.20μg/L;精密度、稳定性、重复性试验的RSD均小于3%;加样回收率为97.08%~106.65%(RSD为0.64%~2.33%,n=9);林下西洋参含量为0.288 2~148.600 0μg/g,园地栽培西洋参含量为0.135 7~65.190 0μg/g。结论:该方法灵敏度高、准确性好,可用于同时测定林下西洋参和园地栽培西洋参中14种微量元素的含量;林下西洋参和园地栽培西洋参中14种微量元素含量差异显著,总体上林下西洋参较园地栽培西洋参中微量元素的含量更高,表明两种生长环境对西洋参的成分影响较大。 OBJECTIVE: To establish a method for simultaneous determination of 14 kinds of microelements from underwood-cultivated Panax quinquefolius and garden-cultivated P. quinquefolius. METHODS:ICP-MS method was adopted. The radio frequency power was 1 350 W,and sampling depth was 7.0 mm;carrier gas velocity was 1.1 L/min,and atomizing chamber temperature was 2 ℃;peristaltic pump was 0.1 rps;plasma gas flow rate was 15.0 L/min in high-salt nebulizer as atomizer;oxide index(CeOZ~+/Ce~+)is less than 0.5% and the double charge index(Ba2+/Ba+)is less than 1%. The microware digestion power is 3 600 W,and the heating process is heated to 190 ℃,maintained at 48 h,and cooling to room temperature,and the heated 150 ℃,maintained 12 h. RESULTS:The linear range of Mn,Cr,Sr,Zn,Rb,B,Ni,Cu,V,Sn,As,Mo,Se and Co were 0.1-500 μg/L(r=0.999 8),0.5-500 μg/L(r=0.999 9),0.1-500 μg/L(r=0.999 9),0.5-500 μg/L(r=0.999 9),0.5-500 μg/L(r=0.999 9),0.1-200μg/L(r=0.999 9),0.5-500 μg/L(r=0.999 9),1-500 μg/L(r=0.999 9),0.1-500 μg/L(r=0.999 8),0.1-200 μg/L(r=0.999 8),0.5-500μg/L(r=0.999 9),0.1-200 μg/L(r=0.999 8),0.5-500 μg/L(r=0.999 9)and 0.1-500 μg/L(r=0.999 9),respectively. The limits of quantitation were less than 0.60 μg/L,and the limits of detection were lower than 0.20 μg/L. RSDs of precision,stability and reproducibility tests were no more than 3%;the recoveries were 97.08%-106.65%(RSDs were 0.64%-2.33%,n=9);the content of underwood-cultivated P. quinquefolius were 0.288 2-148.600 0 μg/g,the content of garden-cultivated P. quinquefolius were 0.135 7-65.190 0 μg/g. CONCLUSIONS:The method is sensitive,accurate and can be used for simultaneous determination of 14 kinds of microelements. There are great differences in 14 kinds of microelements between underwood-cultivated P. quinquefolius and garden-cultivated P. quinquefolius. On the whole,the contents of microelements in underwood-cultivated P. quinquefolius are higher than garden-cultivated P. quinquefolius. The cultivation environments have great influence on the growth of P. quinquefolius.
