摘要
目的:研究麻黄定喘汤和鹿茸大补汤对昆明小鼠灌胃的急性毒性,并评价其安全性。方法:选取昆明鼠种50只,雄雌各半,灌胃给药。通过预实验结果,半数致死量(LD50)足够大,无明确值,采用临床成人药量的100倍,麻黄定喘汤给药组采用23 000 mg·kg^(-1),鹿茸大补汤给药组采用26 000 mg·kg^(-1),并设置生理盐水对照组,按照小鼠体质量10 ml·kg^(-1)的剂量灌胃给药,连续观察7日,并记录小鼠体态、精神状况、体质量、摄食量等。观察7日后,用颈椎脱臼法将小鼠处死,解剖观察,取脑、肝、脾、肺、肾脏器称重,计算脏器系数。对小鼠组织进行生化检查,计算谷草转氨酶活力(U/gprot)和谷丙转氨酶活力(U/gprot)。将麻黄定喘汤给药组和鹿茸大补汤给药组所得数据与生理盐水对照组进行比较。结果:经口急性毒性实验未见动物有中毒症状,无动物死亡,外观表现正常。通过统计处理,所得数据与对照组比较,无统计学意义(P>0.05)。结论:麻黄定喘汤和鹿茸大补汤安全性较高,临床中可以适量使用。
Objective: To study the acute toxicity of Mahuang Dingchuan Decoction and Lurong Dabu Decoction in mice to evaluate their safety. Methods: 50 Kunming mice were given medications through gavage administration. LD(50) was too large to caculate in the preliminary experiment. 100 times dose of clinical adult dosage was applied to mice,i. e. for Mahuang Dingchuan Decoction,it was 23 000 mg·kg^-1; for Lurong Dabu Decoction,it was 26 000 mg·kg^-1; the control group was given normal saline with dose of 10 ml·kg^-1. Body status,mental state,body mass,and food intake were observed in mice for continuous 7 days. After 7 days of observation,the mice were sacrificed by cervical dislocation method. Samples of brain,liver,spleen,lung,and kidney were weighed to calculate organ coefficient. The tissues of mice were examined by biochemical detection to measure the activities of glutamic-oxalacetic transaminase and glutamic-pyruvic transaminase. The data were compared in the three groups. Results: The acute toxicity test showed no symptoms of poisoning,no animal death,and the mice showed normal appearance. There were no significant differences among three groups( P〉0. 05). Conclusion: Mahuang Dingchuan Decoction and Lurong Dabu Decoction have high safety,which can be both applied in clinic with right amount.
