摘要
目的研究银黄清肺胶囊对幼龄大鼠流感病毒肺炎的保护作用及其机制。方法 21日龄SD大鼠60只,雌雄各半,随机分为6组,即正常对照组,模型组,银黄清肺胶囊低、中、高剂量组和阳性药百蕊胶囊组。除正常对照组外,其余5个组动物经鼻腔接种甲型流感病毒,连续3 d,建立流感病毒感染的肺炎模型。除了模型组和正常对照组外,其余组造模当日开始每日灌胃给予银黄清肺胶囊药液(100、200和400 mg·kg^(-1))或百蕊胶囊药液(600 mg·kg^(-1)),每日1次,连续7 d。观察各组大鼠的一般状态;HE染色观察肺组织病理学改变,ELISA检测肺泡灌洗液中成纤维细胞生长因子-2(FGF2)水平;Western Blot检测肺组织成纤维细胞生长因子受体-1(FGFR1)表达。结果与正常对照组比较,经鼻流感病毒感染的模型组动物饮食进水量显著减少,活动量减少,精神状态差,呼吸频率加快,肺组织灌流液中FGF2含量增加,肺组织中FGFR1蛋白表达显著降低;银黄清肺胶囊(400和200 mg·kg-1)能显著改善幼龄大鼠流感病毒肺炎动物的一般情况,减轻肺组织病理学改变,增加肺组织灌流液中FGF2含量和上调肺组织FGFR1蛋白表达。结论银黄清肺胶囊对幼龄大鼠流感病毒肺炎具有较好的保护作用,其机制与调节肺组织中FGF2/FGFR1通路有关。
Objective To determine the protective effect of Yinhuang Qingfei capsules on influenza virus pneumonia in juvenile rats. Methods Sixty 21-day SD juvenile rats (30 males and 30 females) were randomly divided into 6 groups: a normal group, a model group, a positive Bairui capsule group, and 3 dose groups of Yinhuang Qingfei capsules. Except for the normal group, the other groups were inoculated with influenza virus through nose for 3 days to induce pneumonia. After the first inoculation, Yinhuang Qingfei capsules (100, 200 and 400 mg· kg - 1) and Bairui capsules (600 mg · kg - 1) were orally treated once daily for 7 days in corresponding groups. General conditions of the rats were observed every day. At the end of the experiments, the level of fibroblast growth factor 2 (FGF2) in the lung irrigation fluid was determined, and regular histopathology and protein expression of fibroblast growth factor receptor 1 (FGFR1) in the lung tissue were measured. Results Compared with normal rats, some pneumonia symptoms were observed in the rats treated with influenza virus, including poor appetite, fewer autonomous activities, poor mental state, and shortness of breath. The increased level of FGF2 and the downregulated protein expression of FGFR1 were observed in the model rats. Compared with the model rats, Yinhuang Qingfei capsule groups (400 and 200 mg · kg -1) markedly improved these pneumonia symptoms and histopathological changes, and increased the level of FGF2 and protein expression of FGFR1 in the lung. Conclusion Yinhuang Qingfei capsules have strong protective effect on influenza virus pneumonia in juvenile rats, which may be related to the modulation of the lung FGF2/FGFR1 pathway.
