柱前衍生-反相高效液相色谱测定茶叶中19种氨基酸含量 被引量:9

Determination of amino acids in tea by reversed high performance liquid chromatography

摘要 氨基酸是构成茶叶滋味的重要组成部分,茶汤的品质与其含量和成分的比例有关,为了更好地研究茶汤的品质,实验建立了一种快速检测茶叶中游离氨基酸的氯甲酸-9芴基甲酯(FMOCCl)柱前衍生-反相高效液相色谱(RP-HPLC)分析方法。该方法的色谱条件为:Phenomenex Kinetex C18反相色谱柱(100 mm×2.1 mm,2.6μm);流动相包含2部分,A相为含0.05 mol/L乙酸钠的水溶液(pH5.28),B相100%乙腈,梯度洗脱;流速0.15 mL/min;柱温(35±1)℃;紫外检测波长262nm,进样体积10μL时,在(0.004~1)mg/mL浓度范围内,19种氨基酸质量浓度与峰面积呈良好的线性关系,线性相关系数0.988~0.999,检出限(LOD,S/N≥3)范围(0.4~300)μg/mL,定量限(LOQ,S/N≥10)范围(0.8~500)μg/mL,且平均回收率90.29%~105.38%,相对标准偏差(RSD)范围0.36%~15.38%。应用此方法比较分析了不同香型凤凰单丛茶中19种游离氨基酸含量。 Amino acid is an important component of tea taste. The aim of this work was to establish a method for the quality of tea by pre-column derivatization reversed high performance liquid chromatography(RP-HPLC). The samples were derived by 9-fluorenylmethylchloroformate(FMOC-Cl)and HPLC method. Samples were injected with 10 μL into a reverse phase Phenomenex Kinetex C18 column(100 mm×2.1 mm, 2.6 μm). The column operated at(35±1)℃ with a flow rate of 0.15 mL/min using 50 mmol/L acetate buffer(pH5.28) as eluent A and 100% acetonitrile as eluent B. The detection wavelength was operated at 262 nm. The calibration curves had good linearity over concentration range of(0.004-1)mg/mL. The linear relative coefficient for 19 amino acids over concentration range of(0.004-1)mg/mL were 0.988-0.999. The limit of detectins(LOD, S/N≥3) were(0.4-300)μg/mL,and limit of quantifications(LOQ, S/N≥10) were(0.8-500)μg/mL. The mean recovery ratio were90.29%-105.38% with RSDs of 0.36%-15.38%. The 19 free amino acids in different flavor Fenghuang single tea(a kind of Oolong tea) were analyzed by this method.
