摘要
目前甜菜DNA提取通常使用CTAB法,大多数人仍采取研钵研磨的方法,但是在大批量提取DNA的时候,该方法耗时费力,提取效率较低。为了提高提取DNA的效率以适应大规模PCR的需求,我们创新了使用CTAB提取DNA的方法。该方法为先在离心管放入钢珠,然后把样品放入离心管中,放入到液氮中冷冻后直接使用震荡研磨机研磨后,再利用CTAB法进行提取,结果表明,该法点提取的DNA浓度和纯度均表现较好,并且将提取的DNA用于SSR引物的扩增,所得条带清晰,实验结果较好,可提高效率30%。
At present, the CTAB method is usually used for the extraction of sugar beet DNA. Most people stilladopt the method of artificial grinding with mortar, but when extracting DNA in large quantities, the method istime-consuming and labor-intensive, and the extraction efficiency is low. In order to increase the efficiency ofDNA extraction to meet the needs of large-scale PCR, we have innovated one method for extracting DNA usingCTAB. Firstly, the steel ball is placed into the centrifuge tube, then the sample is loaded into the centrifuge tubeand frozen in liquid nitrogen, next directly ground using an oscillating grinder, and then extracted by the CTABmethod. The results show that the DNA concentration and purity extracted by the method are obtained with goodperformance, the extracted DNA is used for amplification of SSR primers, the obtained bands are clear, and theexperimental results are good, which can increase efficiency by approximately 30%.
