摘要
目的:研究衰老对小鼠肠黏膜上皮屏障结构与功能的影响。方法:雄性SPF级C57BL/6小鼠按发育阶段的不同分为1月龄组、6月龄组、10月龄组、12月龄组。采用HE染色、PAS(高碘酸-Schiff试剂)染色、甲苯胺蓝染色对小肠黏膜形态与结构进行观察测量并统计免疫细胞数量;免疫组织化学染色检测紧密连接蛋白(Occludin)的表达。结果:随着月龄增加,小鼠小肠各段绒毛长度均呈降低趋势,相同月龄不同肠段相比,绒毛长度从十二指肠到空肠、回肠呈递减趋势。十二指肠绒毛宽度呈先增高后降低趋势,空肠和回肠绒毛宽度呈下降趋势。十二指肠隐窝深度随月龄增高呈增高趋势,空肠和回肠的隐窝深度随月龄增长呈先增高后降低趋势。随月龄增长,小鼠十二指肠、空肠、回肠各肠段中上皮内淋巴细胞数和肠壁固有层肥大细胞呈先增高后降低趋势,杯状细胞数呈下降趋势。小肠上皮黏膜组Occludin表达随着月龄升高逐渐减少。结论:衰老过程中,小鼠小肠形态发生了一系列结构性改变,黏膜组织中免疫细胞数量减少,肠黏膜上皮屏障免疫相关功能下降。
Objective: To observe the dynamics of intestinal mucosal epithelial parameters during the aging development in mice. Methods: Male SPF C57 BL/6 mice were divided into four groups: 1 month group,6 months group,10 months group and 12 months group according to their developmental stages. HE staining,PAS(periodate-Schiff reagent) staining,toluidine blue staining were used to observe the morphological structure of small intestinal mucosa as well as observe and counted the number of immune cells and the changes of alkaline phosphatase,immunohistochemical staining for expression of tight junction protein(Occludin). Results: With the aging development,the villus height in each segment of the mice's small intestine showed a decreased trend,compared to the same intestine of different months ages,the length of the villus from the duodenum to the jejunum and ileum showed a decreasing tendency. The width of the villus of the duodenum showed a tendency of increasing first and then decreased,and the width of the villus of the jejunum and ileum showed a decreasing trend. The depth of duodenal crypts showed an increasing trendy with the age,and the crypt depth of jejunum and ileum increased at first and then decreased. With the aging development,the number of intraepithelial lymphocytes and the mast cells in the intrinsic layer of the intestinal wall in the duodenum,jejunum,and ileum of mice increased at first and then decreased,the number of goblet cells showed a decreasing tendency. The expression of Occludin in the epithelial mucosa of the small intestine gradually decreased with the aging development. Conclusion: During the aging process,a series of structural changes occurred in the small intestine of mice,the number of immune cells in the mucosal tissue decrease,and the immune-related function of intestinal mucosal epithelial barrier decreased as well.
