MRS评价前列腺癌裸鼠皮下移植瘤CIK治疗疗效的研究 被引量:1

The value of MRS in assessing immunotherapy effect of CIK in human prostate colon cancer transplanted subcutaneously in nude mice

摘要 目的研究磁共振波谱成像技术(MRS)评价CIK细胞对人型前列腺癌裸鼠皮下移植瘤免疫治疗的疗效。方法选取48只裸鼠建立人型前列腺癌PC-3皮下移植瘤模型,随机分为CIK免疫治疗组(n=24)和空白对照组(n=24),CIK免疫治疗组裸鼠每天在接种部位注射0.2mL 1×10^(10)/L的CIK细胞,空白对照组裸鼠每天腹腔注射等量生理盐水,2组分别于治疗前、治疗第7、14、21d进行MRS扫描,CIK免疫治疗组在治疗后的每个时间节点磁共振扫描结束后分别处死8只裸鼠,取肿瘤组织进行常规病理检查及TUNEL凋亡检测。观察MRS获得的反映肿瘤代谢水平的参数值(枸橼酸盐Cit、肌酸Cre以及胆碱Cho)并分析(Cho+Cre)/Cit比值,并与同期肿瘤组织TUNEL凋亡检测结果进行Spearman相关性分析。结果治疗前,空白对照组与CIK免疫治疗组裸鼠的(Cho+Cre)/Cit比值的差异无统计学意义。与空白对照组比较,在治疗后的第7、14、21d,CIK免疫治疗组Cit峰高逐渐升高,Cho峰逐渐降低,(Cho+Cre)/Cit比值逐渐降低,差异均有统计学意义(P <0.05)。肿瘤组织TUNEL凋亡检测结果显示,CIK免疫治疗组与空白对照组比较前列腺癌细胞的凋亡增加。对2组获得的MRS参数与肿瘤组织TUNEL凋亡检测进行相关性分析,结果表明CIK免疫治疗组及空白对照组进行治疗后,3个不同时间节点的(Cho+Cre)/Cit值与肿瘤细胞凋亡检测结果呈负相关(P均<0.05)。结论 CIK细胞免疫治疗对人型前列腺癌移植瘤具抑制作用,能够加快前列腺癌细胞的凋亡。MRS成像技术获得的参数Cit、Cho及(Cho+Cre)/Cit比值能够直观准确地反映前列腺癌治疗前后癌细胞的代谢水平,对于评价前列腺癌CIK免疫治疗的效果具有很大的应用价值。 Objective To evaluate the effect of magnetic resonance spectroscopy (MRS) on the immunotherapy effect of CIK cells on human subcutaneous allograft of prostate cancer in nude mice. Methods A total of 48 human prostate cancer models of PC-3 in nude mice were established, and were randomly divided into CIK immunotherapy group and control group, with 24 of each group. CIK immunotherapy group were injected CIK cells with dose 0.2ml 1×10^10 /L one time and the control group were injected with the same amount of 0.9% sodium chloride so-lution in the same locations, MRS scans were performed before therapy and 7 days,14 days, 21 days after treatment. After MRI scanning, 8 mice were sacrificed respectively in three time points,and the tumor tissue were subjected to pathological examination and TUNEL examination. The following parameters (Cit, Cho and Cre) reflecting the tumor metabolism level were observed and (Cho+Cre)/Cit was analyzed. The correlations between the changes of tumor cells apoptosis and (Cho+Cre)/Cit parameter at each designed time point were analyzed. Results No statistically difference was observed between (Cho+Cre)/Cit of the two groups before treatments. Two groups of 7 days, 14 days, 21 days scan results show that CIK immunotherapy group Cit peak rise gradually, Cho peak gradually decreased, however the control group Cit peak gradually reduce, Cho peak gradually rise, the (Cho+Cre)/Cit ratio reduce, the statistical differences were significant (P 〈0.05). Compared with the control group, the apoptosis of tumor cells were increased in CIK immunotherapy group. the (Cho+Cre)/Cit ratio of the two groups after treatments negatively correlated with tumor cells apoptosis at each time point (P 〈0.05). Conclusion CIK immunotherapy has a better inhibitory effect on human prostate cancer transplantation tumor and can accelerate the apoptosis of tumor cells. The parameters obtained by MRS imaging can directly reflect the metabolic level before and after treatment of prostate cancer, and it is of great value to evaluate the efficacy of CIK immunotherapy in prostate cancer.
