吡格列酮联合克罗米酚治疗PCOS对胰岛素信号转导、氧化炎症反应的影响 被引量:9

摘要 目的:吡格列酮联合克罗米酚治疗多囊卵巢综合征(PCOS)对胰岛素信号转导、氧化炎症反应的调节作用。方法:选择在本院诊断为多囊卵巢综合征的患者作为研究对象,随机分为两组,实验组接受吡格列酮联合克罗米酚治疗、对照组接受克罗米酚治疗;治疗前及治疗后3个月经周期时,测定外周血中胰岛素信号转导分子的表达量、血清中氧化应激及炎症反应指标的含量。结果:治疗后3个周期时,两组患者组内外周血中IRS-1、IRS-2、ERK2、AKT2、GLUT4的表达量以及血清中SOD、TOS、TAS、Adropin、IL-10的含量均显著高于治疗前,血清中RNS、ROS、MDA、PTX3、Apelin、TNF-α的含量均显著低于治疗前且实验组患者治疗后外周血中IRS-1、IRS-2、ERK2、AKT2、GLUT4的表达量以及血清中SOD、TOS、TAS、Adropin、IL-10的含量均显著高对照组,血清中RNS、ROS、MDA、PTX3、Apelin、TNF-α的含量均显著低于对照组。结论:吡格列酮能够通过调节胰岛素信号转导、氧化炎症反应协同克罗米酚治疗PCOS。 Objective: To study the effect of pioglitazone combined with clomiphene treatment of polycystic ovarian syndrome (PCOS) by regulating the insulin signal transduction and oxidative inflammatory response. Methods: The patients who were diagnosed with PCOS in People's Hospital of Xishui County between March 2014 and October 2017 were chosen as the research subjects and randomly divided into two groups: experimental group received pioglitazone combined with clomiphene treatment, and control group received clomiphene treatment. The expression levels of insulin signal transduction molecules in peripheral blood as well as the contents of oxidative stress and inflammatory response indexes in serum were measured before treatment and 3 menstrual cycles after treatment. Results: At 3 menstrual cycles after treatment, peripheral blood IRS-1, IRS-2, ERK2, AKT2 and GLUT4 expression levels as well as serum levels of SOD, TOS, TAS, Adropin and IL-10 of patients in both groups were significantly higher whereas serum levels of RNS, ROS, MDA, PTX3, Apelin and TNF-α were significantly lower than those before treatment. Peripheral blood levels of IRS-1, IRS-2, ERK2, AKT2 and GLUT4 as well as serum levels of SOD, TOS, TAS, Adropin and IL-10 of experimental group after treatment were significantly higher whereas serum levels of RNS, ROS, MDA, PTX3, Apelin and TNF-α were significantly lower than those of control group. Conclusion: Pioglitazone combined with clomiphene can treat PCOS by regulating insulin signal transduction and oxidative inflammatory response.
