老年大鼠脑缺血神经元线粒体形态计量学研究

A morphometric study of neuronal mitochondria in the border zone of the cerebral ischemia induced photochemically-effect of age

摘要 目的 探讨年龄对光化学诱导的局灶性脑缺血皮层神经元线粒体形态结构的影响。方法 对老年和青年大鼠缺血后 4小时、2 4小时、5天缺血边缘区行电镜观察 ,通过计算机图象分析系统对神经元线粒体进行形态计量分析。结果 老年组神经元线粒体数密度和比表面较青年组小 (P <0 .0 5~ 0 .0 1)、而平均体积增大。在相同时间点的缺血边缘区 ,老年组神经元线粒体平均体积、圆球度均较青年组大 ,而比表面减小。两组线粒体平均体积、圆球度及体密度在缺血 4小时增加、2 4小时更明显 ,5天后减小 ,比表面的变化则相反(P <0 .0 5 )。结论 由于老年大鼠存在线粒体老化等年龄相关性改变 ,在同等缺血打击下 ,线粒体肿胀较青年大鼠更严重 。 Objective To examine the effect of age on mitochondria structure of cortical neurons in a photochemical focal cerebral ischemia model. Methods Cerebral ischemia is induced photochemically according to the procedure described by Waston. By electronmicroscpoic examination on the control and the border zone of the cerebral ischemia at the time of 4 hours, 24 hours and 5 days after ischemia induction in young and aged rats, morphometric quantitative study of neuronal mitochondria was investigated with colour pathological image analysis system. Results Compared with young rats, the neuronal mitochondrial numerical density and specific surface were lower( P < 0.05 ) and average volume higher( P < 0.05 ) in aged rats before ischemia. The mitochondrial average volume and sphericity increased and specific surface decreased significantly in ischemic borderline in aged rats as compared to those in young rats at the time of 4 hours, 24 hours and 5 days after ischemia induction( P < 0.05 ). Conclusion The present study indicates that, because of the age dependent changes such as mitochondrial aging, in spite of similar ischemic condition, the mitochondria swelling is more significant and the repair capability lower with respects to young rats.
