睾丸组织原位三维测量生精细胞核的DNA含量与倍体 被引量:14

摘要 细胞(核)的化学成份含量,应以单个完整细胞(核)的测量为基础。薄组织切片内的细胞(核)化学成份含量只是其完整细胞(核)化学成份含量的一部分,不能用于不同细胞(核)群体间的比较。本文使用TIGER细胞图像分析仪,以睾丸组织生精细胞核的三种DNA含量与倍体(单倍体、二倍体和四倍体)关系为模型,探索在组织切片原位测量和计算以单个完整细胞(核)体积为单位的化学成份含量的新方法。结果如下:(1)在无近似圆球形早期精子细胞核的曲细精管切面内,初级精母细胞核的DNA含量约为精原细胞核DNA含量的两倍;(2)在有近似圆球形的早期精子细胞核的曲细精管切面内,精原细胞核的体积明显变小,初级精母细胞核的DNA含量约为近似圆球形早期精子细胞核的四倍,初级精母细胞核与精原细胞核DNA含量的比率大于2,而精原细胞核与早期精子细胞核DNA含量的比率小于2,可能与这段曲细精管内的精原细胞核体积变小,染色质浓缩,使其对Feulgen的着色性降低有关;(3)在4μm薄切片上,采用与测量细胞学样品同样的方法,不能获得如上有规律性的结果。本文结果说明,在组织切片上采用恰当的抽样与测量计算方法,能较好地利用组织切片内细胞(核)的三维形态信息,获得以? Because of the chemical components of cells or nucleir will be partially lost artifactually on thinslices, the data investigated from routine tissue sections can not reflex the true feature of intact cells or nuclei. In this study, a nove method of measurement and analysis strategy with TIGER cell image analysis system was designed to obtain the DNA content in volume unit of whole nuclei in rat spermatogonia, primary spermatocytes and early spermatids of the seminiferous tubules, which containing three types of DNA content and ploidy. Our results showed (1) On the cross sections of seminiferous tubules,the DNA content of the non-global primary spermatocytes was about two times as much as that of spermatogonia; (2) On the cross sections of seminiferous tubules,the DNA content of primary spermatocytes was about four times as much as that ofthe global early spermatids, and the DNA content ratio of primary spermatocyte to spermatogonia was more than 2,but that of the spermatogonia to the early spermatids was less than 2, (3) On the 4μm tissue sections, the same results as above could not be obtained by using the same method on the cytological slices. Conclusion: The method recommended by this paper may be suitable to acquire the chemical composition contents in volume unit of whole cell or nucleus on tissue sections.
