摘要
用基因工程表达并纯化的版纳猪SLAI类P1为抗原 ,经两轮刺激PBMC以3H TdR法检测 1 0名健康人外周血中的特异性T细胞前体频率在 6 67× 1 0 7～ 1 0 8× 1 0 6 之间。用ELISA与RT PCR法进一步分析了抗原特异性细胞的细胞因子分泌特点 ,发现所有抗原阳性孔都呈现IFN γ (+) /IL 4( )的典型Th1型格局。以上结果部分明确了外周血SLA P1特异性T细胞的特征 ,为揭示SLA P1特异性T细胞在猪
Using recombinant Banna Pig SLA class I P1 molecules as antigen,with two rounds of stimulation in vitro,the frequencies of the precursor T lymphocytes in PBMC from 10 healthy individuals were identified,ranging from 6 67×10 -7 to 1 08×10 -6 In a meanwhile,the cytokine profiles of SLA P1 specific T lymphocytes were assayed using ELISA and RT PCR All antigen positive wells showed the typical Th1 type cytokine profile,i e IFN γ (+)/IL 4 ( ) The results partly identified in vitro the characteristics of SLA P1 specific T lymphocytes in healthy PBMC,indicating that SLA P1 specific T lymphocytes were possibly responsible for xeno acute cellular rejection
