摘要
采用聚氨酯类材料为载体固定化根霉BUCT 11发酵生产脂肪酶。对根霉BUCT 11游离脂肪酶催化反应的工艺条件进行了优化 ,采用双温程等手段以后 ,产物中单甘酯质量分数可达 4 7 5 %。固定化细胞简单处理后 ,用于甘油解反应 ,在最佳条件下双温程后 ,单甘酯质量分数达到 35 5 %。文中实现了根霉BUCT 11发酵产物的综合利用 ,降低了单甘酯合成中酶的成本 ,因而具有良好的工业应用前景。
Lipase was produced by immobilized Rhizopus BUCT 11 with polyurethane. Both the lipase and immobilized cell were used for synthesis of monoglyceride. Among several lipases from different sources the lipase of Rhizopus BUCT 11 was found to be suitable for synthesis of monoglycerides. The optimization of glycerolysis conditions of palm oil by free Rhizopus BUCT 11 lipase was made. Under the optimal conditions(enzymatic activity: 315?U/g palm oil, temperature: 30?℃, water content in glycerol: 4 00% and reaction time: 24?h) the MG yield achieved 37 4%, and by a two step temperature reduction the monoglyceride content increased to 47 5%. After simply pretreated the immobilized cells were also used to catalyze the glycerolysis reaction of palm oil. By study of the glycerolysis activity of the immobilized cells, the optimal condition (temperature: 28?℃, cell added: 0 6?g/g palm oil, water content in glycerol: 3 00% and reaction time: 36 h) was attained under which the MG yield arrived at 29 0%, and by a two step temperature reduction 35 5% monoglyceride was achieved.
