摘要
目的 分析新疆地区细粒棘球蚴 95 (EG95 )基因结构特点。 方法 从新疆地区细粒棘球蚴原头节中提取基因组 DNA,以细粒棘球蚴原头节 DNA为模板进行 PCR扩增 ,获得 EG95基因 ,将其克隆至 p UCm- T载体 ,利用 DNAman软件及 Gene Bank/BL AST功能测序并对其进行基因全序列分析。 结果 测序显示新疆株 EG95 (EG95 - XJ)基因片段为 1191bp。 BL AST比对分析表明 EG95 - XJ基因与新西兰株 EG95基因家族成员同源性达 86 %~ 97% ,所有的碱基差异均发生于内含子区域。 结论 中国新疆地区的 EG95基因为 EG95基因家族成员之一 ,但其序列与新西兰株 EG95基因之间存在一定的差异。
Objective To study the structure specificity of Echinococcus granulosus 95(EG95) gene in Xinjiang, China. Methods The genomic DNA was extracted from the protoscolex of E. granulosus in Xinjiang. EG95 gene was amplified by PCR from E. granulosus genomic DNA and was cloned into pUCm T plasmid, and then sequenced. The sequences were analyzed by DNAman and GeneBank/BLAST biosoftware. Results DNA sequence analysis of EG95 Xinjiang strain (EG95 XJ) genomic fragment indicated that the length of EG95 XJ was 1 191 bp. By homology comparison, there were 86%~97% homology of EG95 gene between EG95 XJ and other EG95 gene family members of New Zealand strain, genomic sequence and structure of the EG95 gene is conserved. However EG95 XJ is different from them, the multiple nucleotide differences which are present in EG95 XJ gene in comparison to that of the New Zealand strain occur predominantly in the non coding regions of gene. Conclusion EG95 XJ belongs to the same EG95 gene family, but there is a sequence difference between the EG95 XJ and that of the New Zealand strain.
