摘要
目的 研究不同批号蕲蛇酶静脉给予家兔后 ,对血液凝固系统各项指标的影响 ,以判定其药效的稳定性。 方法 取家兔 ,纯白色 ,体重 1.5~ 2 .5kg,雌雄兼用。每日静脉注入蕲蛇酶 1.0 u/kg.d- 1,连用 3~ 5天 ;药前、药后从心脏取血 6 m l,取全血 1m l作血栓形成 ;余血以 3.0 %枸橼酸钠按 1:9抗凝 ,分离富血小板血浆 ( PRP) ,测定其血小板数目和血小板聚集率。以贫血小板血浆 ( PPP)测定凝血酶时间( TT)、凝血酶原时间 ( PT)、部分凝血活酶时间 ( KPTT)以及纤维蛋白原 ( Fg)含量。 结果 5个批号的蕲蛇酶药后 3天或 5天均可明显使血栓形成的重量减轻 ,长度缩短 ,Fg含量减少 ( P <0 .0 1) ,TT、PT、 KPTT均延长 ( P <0 .0 5~ 0 .0 1) ;血小板数轻度减少 ,聚集率抑制达 2 0 %~ 50 %。 结论 不同批号的蕲蛇酶静脉给予家兔 ,具有显著的作用于血液凝固系统 ,导致血栓形成减少 ,表明对心脑血管内血栓形成药效学相同 。
Objective Studies on the biological activities of various batches of the acutobin on rabbit's blood coagulation system for evaluating the effectiveness of the drug Methods White color rabbits,1 5~2 5 kg were used; Acutobin 1 0U/kg d iv for 3~5 days consecutive Before and after the acutobin was given, blood of 6ml samples were drawn by heart puncture First, 1ml blood for thrombosis determined, then the remain blood ( 5ml ) were prepared for PRP by adding 3 0% sodium citrate(1:9) and centrifuging The Platelets number and aggregation of platelets were determined Thrombin time ( TT ), prothrombin time (PT), Kaolin partial thromboplastin time (KPTT) and fibrinogen level of the PPP were determined Results After intravenous injected the five batches of acutobin for 3~5 days respectively, all the experimental animals of five groups showed same positive effects The thrombosis and fibrinogen level were diminished The TT, PT and KPTT were prolong The platelets number minimal decreased and the aggregation of platelet was inhibited Conclusion Applicaton of rabbits to assay the biological effects of acutobin is necessary and practical
