摘要
紫外可见分光光度法测定桑叶总黄酮含量,并采用正交试验对桑叶中总黄酮的提取工艺进行优化,确定了采用丙酮-水系统溶剂、超声波提取方式提取桑叶中总黄酮的最佳提取工艺。实验结果表明:桑叶中总黄酮的含量为2.78%。丙酮-水系统超声波提取的最佳工艺参数是丙酮浓度为50%、超声波温度70℃、超声时间为40min、料液比为1:20。在此工艺条件下,桑叶总黄酮的提取率最高可达98.6%,RSD为0.22%,其吸光度值在2h内基本稳定。此方法重复性好,简单、易于操作。
Total flavonoids from mulberry leaves were determined in this paper, Optimization of extraction process of totalflavonoids in mulberry leaves by orthogonal test. The acetone-water system solvent, ultrasonic extraction to extract the bestextraction technology of total flavonoids in mulberry leaves. The test results showed that the content of total flavonoids frommulberry leaves was 2.78%. The optimum process paramenters were acetone concentration of 50%, ultrasonic temperature70益, ultrasonic time 40min, ratio of solid to liquid was 1:20. Under these conditons, the recovery rate of total flavonoidsfrom mulberry leaves is 98.6%, RSD 0.22%, the absorbance was stable within 2h. This method good reproducibility, simple,easy to operate.
