摘要
目的研究白芍总苷(TGP)对大鼠动脉粥样硬化的保护作用并初步探讨其作用机制。方法120只SD大鼠随机分为正常对照组、模型组、TGP[50、100、200 mg/(kg·d)]组和辛伐他汀[SV,1.8 mg/(kg·d)]组,每组20只,除正常对照组外,其余组均采用高脂饲料喂养结合腹腔注射维生素D3(VD3)的方法建立SD大鼠动脉粥样硬化模型,造模过程分别同步灌胃给药,共12周。称量心脏质量并计算心脏指数(CI),采用H&E染色法观察各组大鼠主动脉形态结构改变并进行病变分级;生化分析法测定血清中血脂监测指标(TC、TG、LDL-C、HDL-C);比色法测定血浆中炎症细胞因子(CRP、TNF-α、IL-1β、IL-6、IL-10)含量水平和血清中抗氧化酶(SOD、GSH-Px、CAT)活性及丙二醛(MDA)含量。结果较模型组,TGP干预组动脉粥样硬化大鼠CI显著降低,主动脉病变明显改善,TGP[100、200mg/(kg·d)]组病变分级水平显著降低,血清中TC、TG、LDL-C含量显著降低,血浆中CRP、TNF-α、IL-1β、IL-6含量明显降低,血清中SOD、CAT活性显著升高且MDA含量显著降低;TGP 200 mg/(kg·d)组血清中HDL-C含量和GSH-Px活性菌显著升高,血浆中IL-10含量显著升高;上述差异均具有统计学意义(P<0.05,P<0.01)。结论 TGP对大鼠动脉粥样硬化具有一定的保护作用,其作用机制可能与TGP能够有效调节血脂、抑制炎症和降低氧化应激损伤有关。
Objective To investigate the protective effect and mechanism of total glucosides of paeony on atherosclerosis in rats. Methods 120 SD rats were randomly divided into control group, model group, TGP group(50 mg/kg·d), TGP group(100 mg/kg·d), TGP group (200 mg/kg·d) and simvastatin group (1.8 mg/kg·d, SV group), 20 rats in each group. Except the control group, the other groups were treated with high fat diet combined with intraperitoneal injection of vitamin D3 (VD3) to establish SD rat atherosclerosis (AS) model. During the modeling process, the rats were given intragastric administration for 12 weeks. The cardiac quality was measured and the cardiac index (CI) was calculated. The morphological changes of the aorta were observed by H & E staining.The blood lipid monitoring indexes (TC, TG, LDL-C (IL-1β, IL-6, IL-10), serum antioxidant enzymes (SOD,GSH-Px, CAT) and malondialdehyde (MDA) were measured by colorimetry. were measured by colorimetry. Results Compared with the model group, CI in TGP groups were significantly decreased, the aortic disease was significantly improved. The levels of CRP, TNF-α, IL-1β and IL-6 in plasma were significantly decreased (P<0.05), and the levels of TC, TG and LDL-C were significantly decreased; the activity of SOD and CAT in serum was significantly increased and the content of MDA was significantly decreased in the TGP group(100 mg/kg·d) and TGP group(200 mg/kg·d). The content of HDL-C and the activity of GSH-Px in TGP group 200 mg/(kg·d)were significantly increased and the content of IL-10 in plasma was significantly increased (P<0.05, P<0.01).Conclusion TGP has protective effects on atherosclerosis rats, which perhaps related to its effects of reducing cholesterol, improving the body's anti-inflammatory and antioxidant capacity.
