摘要
目的本文旨在系统评价FasL-844C/T基因多态性与乳腺癌易感性之间的关系。方法计算机检索PubMed、EMBASE、中国生物医学文献数据库(CBM)、中国期刊全文数据库(CNKI)和万方数据库,检索建库至2017年5月8日之间发表的有关FasL-844C/T基因多态性与乳腺癌易感性的病例-对照研究,采用STATA 13.0软件进行Meta分析,计算合并比值比(OR)及其95%可信区间(95%CI)。结果共纳入11个病例-对照研究,包括5698例乳腺癌患者,5710例健康对照。Meta分析结果表明,在杂合子模型(OR=0.789,95%CI:0.651~0.955,P=0.015)和显性遗传模型(OR=0.810,95%CI:0.666~0.986,P=0.036)比较下,FasL-844C/T基因多态性与乳腺癌易感性有显著关联;按种群进行的分层分析发现,亚洲人群,尤其是中国人群FasL-844C/T基因多态性与乳腺癌易感性有显著关联,但欧洲人群FasL-844C/T基因多态性与乳腺癌易感性之间未达到显著性水平。结论 FasL-844C/T基因多态性与乳腺癌易感性之间具有显著关联,尤其在中国人群中T~*突变体具有减少罹患乳腺癌的风险。
Objective This study aims to systematically evaluate the relationship between FasL-844 C/T gene polymorphism and susceptibility of breast cancer. Methods A computer-based online search of the PubMed,EMBASE,Chinese Biomedicine database(CBM),China National Knowledge Infrastructure(CNKI)and Wan Fang Data was carried out.Case-control studies about the relationship between FasL-844 C/T gene polymorphism and susceptibility of breast cancer were collected from the establishment of databases to May 8,2017.Meta-analysis was performed by STATA 13.0 software and the pooled odds ratio(OR)and its 95% confidence interval(95%CI)were calculated. Results A total of 11 case!control studies were included in the Meta-analysis,including 5698 breast cancer patients and 5710 healthy controls.Meta-analysis showed that the FasL-844 C/T gene polymorphism in heterozygous model(OR =0.789,95%CI :0.651~0.955,P =0.015)and dominant genetic model(OR =0.810,95%CI :0.666~0.986,P =0.036)were significantly associated with susceptibility of breast cancer.The stratified analysis based on population showed that the FasL-844 C/T gene polymorphism of Asian population,especially the Chinese population was significantly related with the susceptibility of breast cancer.However,FasL-844 C/T gene polymorphism of Europeans had no significant relationship to the susceptibility of breast cancer. Conclusion FasL-844 C/T gene polymorphism is significantly associated with susceptibility of breast cancer.FasL-844 C/T gene polymorphism T~* mutant is associated with lower risk of breast cancer,especially in Chinese population.
