石上柏抗肿瘤活性与其红外指纹图谱的谱效研究 被引量:12

The relationship between anti-tumor effect of Selaginella Doederleinii Hieron and its infrared fingerprint

摘要 目的探讨中药材石上柏的抗肿瘤活性与其红外指纹图谱之间的相互关系。方法以30批来自不同产地的石上柏为研究对象,采用CCK8试剂检测石上柏乙酸乙酯提取物对人肺癌A-549细胞和人肝癌SMMC-7721细胞的生长抑制效果;傅里叶变换红外光谱仪采集石上柏乙酸乙酯提取物的指纹图谱。根据肿瘤细胞的抑制率和红外指纹图谱特征峰强度采用后退法构建石上柏抑制A-549和SMMC-7721两种肿瘤细胞株的谱效数学模型。结果不同产地的石上柏乙酸乙酯提取物主要含有萜类和双黄酮类化合物;石上柏乙酸乙酯提取物能够抑制A-549和SMMC-7721细胞的生长,当浓度为200μg/m L作用48 h,对细胞的抑制率在50%~90%;所建立数学模型的预测值与实际测量值的偏差率全部在±10%以内。结论石上柏乙酸乙酯提取物在体外对肺癌A-549细胞和肝癌SMMC-7721细胞具有抑制作用,其抗肿瘤活性与石上柏红外指纹图谱之间具有一定的相关性。 To investigate the relationship between the antitumor effect of S.doederleinii Hieron and its infrared fingerprints.Methods30batches of S.doederleinii Hieron from different habitats in China were used.Ethyl acetate extracts of S.doederleinii Hieron were applied against human lung cancer A-549cells and liver cancer SMMC-7721cells and their infrared fingerprints were collected by Fourier Transform infrared spectroscopy.Screening arguments of backward elimination method was used to establish the spectra-effect correlation model between tumor inhibition and infrared fingerprints.ResultsThe main compounds of ethyl acetate extracts of S.doederleinii Hieron from different habitats were terpenoids and biflavonoids.The ethyl acetate extracts of S.doederleinii Hieron had inhibitory effects on A-549cells and SMMC-7721cells.When the concentration of extracts was used as200μg/ml for48h,the inhibition rate ranged from50%to90%.According to the spectra-effect correlation model,the deviations of the predictive values from the true values were within±10%for the two kinds of tumor cells.ConclusionThis study suggests that the flavanoid from S.doederleinii Hieron has antitumor effects in vitro and there is correlationship between antitumor effect and infrared fingerprint in S.doederleinii Hieron.
