摘要
【目的】研究瑞香狼毒根的乙醇提取物石油醚、氯仿萃取部位对截形叶螨、二斑叶螨的触杀活性成分.【方法】采用活性跟踪法分析了其中的活性化合物,并用玻片法进行了室内毒力测定,研究了6种单体化合物对两种叶螨的触杀活性.【结果】得到了5种香豆素类活性化合物和1个甾体化合物,通过核磁共振波谱学数据解析,确定6个化合物结构为:β-谷甾醇,伞形花内酯,东莨菪素,瑞香素,西瑞香素,瑞香苷,6种化合物均有一定的杀螨活性,其中化合物(1)、(4)、(6)对截形叶螨、二斑叶螨具有较强的触杀活性,0.80的化合物1、4、6在24h对截形叶螨的校正死亡率分别达到了45.21%、52.54%和76.49%,LC50分别为0.941 7、0.633 0、0.314 6mmol/L;对二斑叶螨的校正死亡率分别达到了53.16%、62.53%和72.24%,LC_(50)分别为0.647 2、0.526 5、0.374 2 mmol/L.【结论】瑞香狼毒根中杀螨活性成分主要是β-谷甾醇、瑞香素、瑞香苷.
【Objective】To isolate and identify the acaricidal compounds extracted from the root of Stellera chamaejasme.【Method】Petroleum ether and chloroform were as extracting agents.Bioactivity-guided method was used for isolation and identification.The contact activity of six identified compounds were tested against Tetranychus truncates and Tetranychus urticae.【Result】One steroid and five coumarins were isolated from petroleum ether fraction and chloroform fraction.The structures of the purified compounds were elucidated on the basis of spectral data and chemical evidences,which were determined asβ-sitosterol(1),umbelliferone(2),pureonebio(3),daphnetin(4),thymerol(5),daphnin(6).The six compounds exhibited significantly acaricides activities against T.truncates and T.urticae.Compounds1,4and6had stronger acaricides activity than others.Under the concentration at0.80mM,the corrected mortality against T.truncates after24hour were45.21%,52.54%and76.49%,with LC50of0.9417,0.6330and0.3146mmol/L,respectively.The corrected mortality against T.urticae with compounds1,4and6after24hours were53.16%,62.53%and72.24%,with LC50of0.6472,0.5265and0.3742mmol/L,resprctively.【Conclusion】The main contact activity compounds from roots of S.chamaejasme against T.truncates and T.urticae areβ-sitosterol,daphnetin,and daphnin.
