七子益肾理冲汤对DOR大鼠卵巢颗粒细胞凋亡的影响 被引量:1

摘要 目的探讨七子益肾理冲汤对VCD致卵巢储备功能低下(diminished ovarian reserve,DOR)大鼠之卵巢颗粒细胞凋亡的影响。方法 60只雌性SD大鼠随机分为空白组、模型组、中药低、中、高剂量组及DHEA组。模型组采用VCD注射制作DOR大鼠模型。观察各组大鼠HE染色卵巢切片中各级卵泡数及黄体数量,及TUNEL法检测各组大鼠卵巢颗粒细胞凋亡率。结果与空白组比较,模型组卵巢原始卵泡数量及生长卵泡数量显著下降(P<0.05);与空白组比较,中药高剂量组及西药组的原始卵泡数量及生长卵泡数量均无显著差异;模型组较空白组卵巢颗粒细胞凋亡率显著上升(P<0.05),经治疗后,中药中剂量、高剂量组及西药组大鼠的颗粒细胞凋亡率均比较模型组明显降低(P<0.05)。结论七子益肾理冲汤可減少卵巢细胞凋亡,修复卵巢原始卵泡储备的损失。 Objective To investigate the effect of Qizi Yishen Lichong decoction on apoptosis of ovarian granulosa cells in DOR rats model.Methods 60 female SD rats was divided into six groups:blank group,model group,CM low-dose group,CM medium-dose group,CM high-dose group and DHEA group.The DOR rat model was established by injecting with VCD.HE staining of ovarian were observed under microscope to count the number of different types of ovarian follicles and corpus luteum;and apoptosis rate of ovarian granulosa cells were tested by TUNEL method.Results Compared with blank group,the number of primordial follicles and the number of growing follicles of model group were significant decreased(P<0.05).Compared with the blank group,there was no significant difference in the number of primordial follicles and the number of growth follicles in the CM high dose group and the western medicine group.The apoptosis rate of ovarian granulosa cells in the model group was significantly higher than that in the blank group(P<0.05).After treatment,the apoptosis rate of granulosa cell in the CM medium dose,CM high dose group and Western medicine group was significantly lower than that of the model group(P<0.05).Conclusion Qizi Yishen Lichong decoction can reduce the apoptosis of ovarian cells and repair the loss of ovarian follicle reserve.
