苦参碱对人子宫内膜癌细胞增殖抑制的作用机制探究 被引量:6

摘要 目的探讨苦参碱对人子宫内膜癌细胞增殖的抑制作用及其机制。方法实验对象为人子宫内膜癌细胞株Ishikawa(常规培养),采用MTT法检测不同浓度苦参碱(0、5μg/ml、10μg/ml、20μg/ml、40μg/ml、80μg/ml、160μg/ml、320μg/ml)作用24 h、48 h后对Ishikawa细胞增殖抑制率,采用流式细胞技术检测苦参碱对Ishikawa细胞周期的影响,采用Western Blot技术检测苦参碱对Ishikawa细胞血管内皮生长因子(VEGF)、核因子κB(NF-κB)p65蛋白表达的影响。结果随着苦参碱药物浓度的增加,对Ishikawa细胞增殖抑制率明显升高,呈一定的剂量依赖性;苦参碱作用24 h IC50为44.655μg/ml,苦参碱作用48h IC50为20.664μg/ml。Ishikawa细胞接受不同浓度苦参碱处理48 h后,G1期细胞百分比的增高情况明显,G2期以及S期细胞百分比降低(P<0.05),且随着苦参碱药物浓度的增加,变化越显著(P<0.05)。不同浓度苦参碱可有效抑制Ishikawa细胞VEGF、NF-κB p65蛋白表达(P<0.05),且随着浓度的增高,Ishikawa细胞VEGF、NF-κB p65蛋白表达越低(P<0.05)。结论苦参碱能够对人子宫内膜癌细胞产生明显的抑制,而且具有一定程度上的剂量依赖性,其可能具有将肿瘤细胞阻滞于G1期的作用机制,抑制VEGF、NF-κB p65蛋白表达。 Objective To investigate and analyze the mechanism of Matrine on the proliferation inhibition of human endometrial carcinoma cells.Methods Human endometrial carcinoma cell line Ishikawa were cultured,MTT method was used to detect different concentrations of matrine(0μg/ml,5μg/ml,10μg/ml,20μg/ml,40μg/m,80μg/ml,160μg/ml,320μg/ml)24 h,48 h on the proliferation rate of Ishikawa cells by matrine,flow cytometry was used to detect the effect on cell cycle in Ishikawa cells,Western Blot technology was used to detect effects of Matrine on the expression of NF-κB p65 and VEGF protein in Ishikawa cells.Results With the increase of the concentration of matrine,the inhibition rate of Ishikawa cell proliferation was significantly increased in a dose dependent manner.The effect of Matrine on 24 h IC50 was 44.655μg/ml and 48 h IC50 is 20.664μg/ml.After Ishikawa cell 48h was treated with different concentrations of matrine,the percentage of G1 phase cells increased,and the percentage of G2 and S phase cells decreased(P<0.05).As the concentration of matrine increased,the change was more significant(P<0.05).Different concentrations of matrine can effectively inhibit the expression of VEGF and NF-κB p65 protein in Ishikawa cells,and with the increase of concentration(P<0.05),the expression of VEGF and NF-κB p65 protein in Ishikawa cells was lower(P<0.05).Conclusion Matrine has a significant proliferation inhibition effect on human endometrial cancer cells,and is dose dependent.The mechanism may be that it can block tumor cells in G1 phase,and inhibit the expression of VEGF and NF-κB p65 protein.
