东北地区玉米穗腐镰孢菌种类鉴定及拟轮枝镰孢菌遗传多样性 被引量:12

Identification of Fusarium species Causing Maize Ear Rot in Northeast China and the Diversity of F. verticillioides

摘要 为明确东北地区玉米穗腐病致病镰孢菌的种类及拟轮枝镰孢菌的多样性,于2015~2016年在东北4个省30个区县进行玉米穗腐病样品的采集,通过形态学及分子生物学方法对分离获得的镰孢菌的种类进行鉴定,并用ISSR分子标记技术对40株拟轮枝镰孢菌进行遗传多样性分析。结果表明:在获得的122株镰孢菌分离物中鉴定出包括禾谷镰孢菌、层出镰孢菌、亚粘团镰孢菌和拟轮枝镰孢菌共4个种,其分离频率分别为4.92%、7.38%、10.66%和77.05%。从区域分布来看,拟轮枝镰孢菌的分布最广,在所有采样县区均有分布。在30个县区采集的玉米穗腐病样品中,有7个县区分离到亚粘团镰孢菌,有5个县区分离到层出镰孢菌,有2个县区分离到禾谷镰孢菌,分别占采集县区样本的23.33%、16.67%和6.67%,说明拟轮枝镰孢菌是东北地区玉米穗腐病的优势致病菌。ISSR-PCR结果表明,筛选出的8条ISSR引物共扩增出45条清晰条带,其中27条为多态性条带,不同引物对供试菌株扩增的条带数不同,引物842扩增的条带最多,引物873扩增的条带最少,多态性条带的比例分别是66.7%和50%。聚类分析结果表明,供试菌株间的遗传相似系数为0.75~1.00,当遗传相似系数为0.97时,供试菌株被全部分开。拟轮枝镰孢菌ISSR类群的划分与其地理来源没有明显的相关性。 To clarify the population structure characteristics of Fusarium species causing maize ear rot in Northeast China and the diversity of F.verticillioides,the kernel samples were collected from 30 counties in 4 northeastern provinces in 2015-2016.A total species of Fusarium were obtained and identified according to the morphology and molecular biology methods,the genetic diversity analysis of 40 strains of F.verticillioides was performed using ISSR molecular marker technique.The results showed 4 species were identified among the 122 isolates of Fusarium,including F.graminearum,F.proliferatum,F.subglutinans and F.verticillioides.The isolation frequencies were 4.92%,7.38%,10.66%and 77.05%.From the perspective of regional distribution,the distribution of F.verticillioides is the most widespread and is distributed in all sampling counties.Of the corn ear rot samples collected in 30 counties,F.subglutinans was isolated in 7 counties,F.proliferatum was isolated in 5 counties,and F.graminearum was isolated in 2 counties,which accounted for 23.33%,16.67%and 6.67%of the collection counties,respectively.The dominant pathogen of maize ear rot was F.verticillioides.The ISSR-PCR results showed that the 8 ISSR primers were selected to amplify 45 clear bands,27 of which were polymorphic bands.The number of bands amplified by different primer pairs for the tested strains was different.The most amplified band was primer 842,The least amplified band was primer 873,the proportion of polymorphic bands was 66.7%and 50%,respectively.The results of cluster analysis showed that the genetic similarity coefficient between the test strains was 0.75-1.00.When the genetic similarity coefficient was 0.97,the test strain was completely separated.There is no obvious correlation between the division of the ISSR group and its geographic location.
