1,25-二羟维生素D3对人卵巢癌HO-8910细胞增殖、凋亡影响的研究 被引量:4

摘要 目的:通过1,25-(OH)_2-VD_3在体外对人卵巢癌HO-8910细胞进行诱导,探讨其在卵巢癌细胞凋亡过程中的作用机制。方法:以体外人卵巢癌HO-8910细胞培养为基础,采用MTT法检测1,25-(OH)_2-VD_3对HO-8910细胞增殖的抑制作用,通过ELISA法、免疫细胞化学法检测P27蛋白表达的改变。结果:MTT结果显示1,25-(OH)_2-VD_3能抑制HO-8910细胞的增殖,并呈时间及剂量效应依赖性(P<0.05)。ELISA结果显示,经诱导的HO-8910细胞P27蛋白表达升高(P<0.05)。结论:1,25-(OH)_2-VD_3能够通过上调P27蛋白的表达,抑制人卵巢癌HO-8910细胞增殖。 Objective:To investigate the effect of 1,25-(OH)2-VD 3 on human ovarian cancer HO-8910 cells in vitro and to explore its mechanism in the process of apoptosis of ovarian cancer cells.Methods:The inhibitory effect of 1,25-(OH)2-VD 3 on the proliferation of HO-8910 cells was detected by MTT assay.The expression of P27 protein was detected by ELISA.Results:MTT assay showed that 1,25-(OH)2-VD 3 inhibited the proliferation of HO-8910 cells in a time-and dose-dependent manner(P<0.05).The results of ELISA and Immunocytochemical method showed that the expression of P27 protein in HO-8910 cells was increased(P<0.05).Conclusion:1,25-(OH)2-VD 3 can inhibit the proliferation of human ovarian cancer HO-8910 cells by up-regulating the expression of P27 protein.
