摘要 目的 研究肝细胞生长因子 (HGF)对体外培养大鼠颌下腺细胞增殖的剂量 效应及时间 效应。方法 采用体外颌下腺细胞培养在含 1 0 %胎牛血清的合成培养液 (DMEM) ,实验组分别加入不同浓度的HGF ,利用噻唑蓝 (MTT)比色法检测细胞的增殖能力 ,研究HGF对颌下腺细胞增殖的影响。结果 在 1 0 %胎牛血清配制的DMEM含HGF(1 .0~ 1 0 0 0 .0 μg/L)可促进培养细胞的增殖 (P <0 .0 1 ) ,且在一定浓度范围内HGF(1 .0、5 .0、1 0 .0、50 .0、1 0 0 .0 μg/L)呈剂量 效应关系 ,最大效应剂量为 1 0 0 .0 μg/L。加入HGF第 3天开始显示出明显的促增殖效应。 结论 HGF在 1 0 %胎牛血清DMEM培养条件下能够促进颌下腺细胞的增殖和分化 。 Objective To investgate the effect of hepatocyte growth factor on rat submandibular gland cell proliferation.Methods Submandibular gland cells were obtained and purified by amylase digestion from the gland of the rat. The epithelial cells were cultured and subcultured in 10% fetal bovine serum and various concentrations of hepatocyte growth factor on Dulbecco minimum essentia medium. MTT colorimetric method was used to assay numeric value and to estimate vital force of the cultured cells. The relations of dose response and time course were studied by MTT colorimetric method.Results Dose response:In Dulbecco minimum essentia medium containing 10% fetal bovine serum,hepatocyte growth factor (1.0 1?000?μg/L) stimulated the proliferation moderately (P<0.01). In a definite range of hepatocyte growth factor (1.0,5.0,10.0,50.0,100.0?μg/L),hepatocyte growth factor had mitogenic effect dose dependently. Time course:The growth curve of experiment group moved to left and flat stage appeared in advance. Cell proliferation study indicated that hepatocyte growth factor had obvious promotion effect on cell proliferation.Conclusion Hepatocyte growth factor can promote the proliferation and differentiation of submandibular gland cells significantly in 10% fetal bovine serum conditions on Dulbecco minimum essentia medium.
