摘要
机体维持内环境的氧稳态是通过调节缺氧诱导因子1α(HIF-1α)的表达实现的,而心脏在维持氧稳态中起着关键作用。心脏自身维持有效的收缩功能也需要有充足的氧气供应。HIF-1α作为低氧敏感因子,低氧状态会抑制HIF-1α的降解,导致HIF-1α与HIF-1β结合形成复合物,进而调控细胞核内相关基因的表达,激活多条信号通路,提高细胞的缺氧适应能力,进而保护细胞。HIF-1α在缺血性心肌病与心力衰竭中通过参与血管张力调节、血管生长因子、葡萄糖摄取以及糖酵解等途径调控氧的传递,增强缺氧组织的存活能力,发挥心肌保护的作用。
The oxygen homeostasis that maintains body′s internal environment is the key to regulating the expression of hypoxia inducible factor 1α(HIF-1α).The heart plays a key role in maintaining the oxygen homeostasis,but the heart itself needs adequate oxygen supply to maintain its effective systolic function.HIF-1αis a hypoxia-sensitive factor,and hypoxia state will inhibit the degradation of HIF-1α.HIF-1αis able to combine with HIF-1βto form a complex,which can regulate the expression of nucleus-related genes,activate multiple signal channels and hypoxia adaptation of cells,thus protecting the cells.HIF-1αparticipates in regulating vascular tension and vascular growth factor,uptaking glucose and glycolysis in ischemic cardiomyopathy and heart failure,so as to regulate oxygen transmission,enhance the survival of hypoxic tissue and protect myocardium.
