摘要
为观察石膏粳米汤对干酵母致热大鼠炎症因子、核因子(NF)-κBp65、环氧合酶-2(COX-2)表达的影响,50只SPF级SD雄性大鼠被随机分正常对照组10只,余40只采用20%干酵母混悬液皮下注射建立大鼠发热模型。4 h后随机分4组:模型组、阿司匹林组(0.1 g·kg^(-1))、石膏粳米汤组(7.83 g·kg^(-1))、石膏组(17.62 g·kg^(-1))。造模后每1 h测量1次大鼠肛温,共10次;计算各组大鼠不同时间点体温变化、最大体温上升高度(ΔTmax)、6 h体温反应指数(TRI6);检测各组大鼠血清TNF-α、IL-1β、IL-6含量;采用免疫组化法、实时荧光定量PCR法检测各组大鼠下丘脑NF-κBp65、COX-2的表达情况。结果是给药后4~6 h内,阿司匹林、石膏粳米汤、石膏水煎液均能抑制发热大鼠体温升高,石膏粳米汤和石膏水煎液之间无明显差异(P>0.05)。给药后6 h,阿司匹林组、石膏粳米汤组、石膏组大鼠血清TNF-α、IL-1β、IL-6浓度均显著低于模型组(P<0.01、P<0.01、P<0.01);三组下丘脑NF-κBp65蛋白、COX-2基因表达均显著低于模型组(P<0.01、P<0.01、P<0.01)。说明石膏粳米汤可改善发热大鼠炎症,其发挥解热抗炎机制可能涉及抑制下丘脑NF-κBp65蛋白、COX-2 mRNA表达相关。
To observe the impact of gypsum rice decoction on antipyretic inflammation,NF-κBp65 and COX-2 in yeast-induced rats,10 from 50 male SD rats randomly were selected to normal control group.The rest of 40 were injected with 20%yeast suspension to establish models of fever.After 4 h of the model,the rats was successfully established fever model.Then the model rats were randomly divided into four groups:10 in model group,10 in aspirin group(0.1 g·kg^-1),10 in gypsum rice decoction group(7.83 g·kg^-1),10 in gypsum decoction group(17.62 g·kg^-1).After injecting yeast,the anal temperature was measured 1 time per 1 h,and 10 times were measured.The temperature of rats at different time points,the maximum temperature rise height(ΔT max)and the temperature response index of 6 h(TRI 6)were calculated.Enzyme-linked immunosorbent assay was used to detect the level of serum TNF-α,IL-1β,IL-6 in each group.Immunohistochemistry and RT-qPCR were used to detect the expression of NF-κBp65,COX-2 in hypothalamus of each group.After administration of 4~6 h,aspirin,gypsum rice decoction and gypsum decoction can inhibit the increase of temperature of febrile rats,showing the antipyretic effect.There is no significant differences between the gypsum rice decoction and gypsum decoction(P>0.05).After administration of 6 h,the level of serum TNF-α,IL-1β,IL-6 in aspirin group,gypsum rice group and gypsum group are all over lower than model group,the difference is statistically significant(P<0.01,P<0.01,P<0.01).The expression level of NF-κBp65 protein and COX-2 gene in hypothalamus in aspirin group,gypsum rice decoction group and gypsum decoction group are all lower than model group,the difference were statistically significant(P<0.01,P<0.01,P<0.01).It is concluded that Gypsum rice decoction has antipyretic effect in yeast-induced rats,the mechanism of antipyretic effect may be related to inhibit the expression of NF-κBp65 protein and COX-2 mRNA in hypothalamus.
