外源ABA喷施提高番茄幼苗抗旱性研究 被引量:11

Study on exogenous ABA on improving drought resistance of tomato seedling

摘要 以番茄栽培品种Moneymaker为试验材料,对四叶期番茄幼苗喷施300μL·L^(-1)脱落酸,干旱处理后测定相关生理指标。结果表明,随干旱处理时间延长,幼苗相对含水量降低,相对电导率升高;同时期抗氧化酶活性及相对含水量均高于CK,丙二醛含量及相对电导率均低于CK。通过cDNA-AFLP技术筛选得到4个与植物抗逆反应相关的差异表达基因片段。实时荧光定量验证表明,干旱胁迫下,两组中4个基因片段(TDF23、 TDF56、TDF62和TDF71)表达量均呈上升趋势。ABA预处理后TDF23和TDF62表达显著高于CK。 Tomato cultivar Moneymaker was used as the experimental material.The four leaf-stage tomato seedlings were sprayed with abscisic acid(300μL·L-1),and the corresponding physiological indices were determined after drought treatment.The results showed that the relative water content of seedlings decreased and relative electrical conductivity increased with the extension of drought treatment time.At the same period,both the activities of antioxidant enzyme and the relative water content were higher than that of CK,while the contents of MDA and the relative electrical conductivity were lower than that of CK.Four differentially expressed gene fragments related to plant stress response were screened by cDNA-AFLP analysis.Real-time quantitative fluorescence verification suggested that the expression levels of four gene fragments(TDF23,TDF56,TDF62 and TDF71)in both groups showed an upward trend under drought stress.The expression patterns of TDF23 and TDF62 after treatment with ABA were significantly higher than that of CK.
