PPAR-γ在脓毒症大鼠肝损伤中的表达及意义 被引量:4

Expression and significance of PPAR-γ in the liver damage in rats with sepsis

摘要 目的:探讨过氧化物酶体增殖物激活受体-γ(peroxisome proliferator-activated receptor-gamma, PPAR-γ)在脓毒症大鼠肝脏中的表达和意义。方法:90只SD大鼠随机等分为假手术组(sham operation group, SO组)、经典盲肠结扎穿刺术组(cecal ligation and puncture group, CLP组)和吡格列酮预处理组(pioglitazone pretreatment group, PIO组),术后3、6、12、24、48 h检测大鼠血中白细胞计数、血清中天门冬氨酸氨基转移酶(aspartate aminotransferase, AST)含量,逆转录聚合酶链式反应(reverse transcription polymerase chain reaction, RT-PCR)检测肝脏组织中PPAR-γmRNA的表达情况,取肝脏组织HE染色作常规病理检查。结果:与SO组比较,CLP组术后肝组织逐渐出现肝细胞水肿、气球样变性、炎症细胞浸润,白细胞计数明显升高,AST含量明显升高,PPAR-γmRNA的表达显著下降。而PIO组的上述指标均明显改善(均P<0.05)。结论:脓毒症大鼠肝脏损伤的发生过程中肝脏细胞PPAR-γ的表达显著下降,使用吡格列酮能有效提高脓毒症大鼠肝脏PPAR-γ的表达,保护肝脏。 Objective:To investigate the expression and significance of peroxisome proliferator-activated receptor-gamma(PPAR-γ)in the liver damage of septic rats.Methods:90 Sprague-Dawley(SD)rats were randomly and equally divided into sham operation group(group SO),operated by cecal ligation and puncture group(group CLP),and pioglitazone pretreatment group(group PIO).At 3,6,12,24,48 h after operation,white blood cell(WBC)count,serum aspartate aminotransferase(AST),the expression of PPAR-γmRNA in liver tissue were mesured by reverse transcription polymerase chain reaction(RT-PCR).Liver tissue′s routine pathological examination was performed by HE staining.Results:Compared with group SO,there was a gradual emergence of hepatocyte edema,ballooning degeneration,inflammatory cell infiltration in the liver tissue of group CLP.WBC,AST increased significantly,while the expression of PPAR-γwas significantly decreased(P<0.05),which were all significantly improved in group PIO(P<0.05).Conclusion:Sepsis decrease the expression of PPAR-γin liver cells,the process of which was involved in the pathogenesis of liver damage in septic rats.Pioglitazone can effectively increase the expression of PPAR-γ,which can protect the liver damage of septic rats.
