摘要
通过利用DNA合成的银纳米簇作为荧光探针研究一种新型的检测汞离子的方法。该方法是基于汞离子触发使颈环DNA的颈部形成双链结构,核酸外切酶Ⅲ剪切双链部分释放出DNA单链,该DNA单链能作为模板用于形成荧光银纳米簇,该传感器的荧光信号与汞离子的浓度有关。释放的汞离子可以继续参与循环,进一步放大检测信号。汞离子检测的线性范围是3.0×10-11到3.0×10-8 mol·L-1,该传感器的灵敏度较高。该方法是对汞离子的特异性检测,不受其他离子的影响,并可用于实际样品的检测。
A novel method for detecting Hg 2+ion was developed using silver nanoclusters as fluorescent probe.The stem-loop structure DNA could form a closed double stranded structure in presence of Hg 2+ions.Then ExoⅢcould recognize and shear the double stranded portion to release DNA single strand.The released DNA single strand acted as a template for the preparation of fluorescent AgNCs.Thus the fluorescence of the AgNCs could be used as a signal transduction of this sensor,which was related to the concentration of the Hg 2+ions.The released target Hg 2+ions could initiate another cycle of the cleavage reaction and leading to signal ampliflcation in the same way.A linear response to the concentration of Hg 2+ions was observed in the range from 3.0×10-11 to 3.0×10-8 mol·L-1.This sensor was specific for Hg 2+ions without being affected by other metal ions.At the same time,it might find important applications for actual sample.
