siRNA沉默HDAC6对宫颈癌Hela细胞顺铂敏感性的影响

摘要 目的探讨小分子干扰RNA(siRNA)沉默组蛋白去乙酰化酶6(HDAC6)对宫颈癌Hela细胞顺铂敏感性的影响。方法选取宫颈癌细胞Hela,分别转染Control siRNA和HDAC6 siRNA,转染后的细胞中分别加入不同浓度的顺铂。采用RT-PCR法检测HDAC6 mRNA表达,Western blot检测HDAC6、Bcl-2、Bax蛋白表达,流式细胞仪检测细胞凋亡,MTT法检测细胞在顺铂作用下的体外存活率。结果 HDAC6 siRNA组HDAC6 mRNA表达(82.3±24.5)低于空白对照组(352.7±89.2)及Control siRNA组(361.7±77.6)(均P<0.05);HDAC6 siRNA组HDAC6、Bcl-2蛋白表达[(118.7±20.9)、(179.5±44.6)]低于空白对照组[(558.4±105.5)、(494.1±65.3)]及Control siRNA组[(564.9±98.5)、(487.2±74.1)](均P<0.05),Bax蛋白表达(448.6±70.8)显著高于空白对照组(189.7±60.3)及Control siRNA组(194.2±68.4)(均P<0.05)。HDAC6 siRNA组细胞凋亡率(37.6±8.7)%,高于空白对照组的(5.8±1.1)%及Control siRNA组的(6.4±1.3)%。不同浓度顺铂作用后,HDAC6 siRNA组IC50值较空白对照组及Control siRNA组显著降低(均P<0.05)。结论沉默HDAC6表达可提高宫颈癌Hela细胞对顺铂的敏感性,而促细胞凋亡可能是其主要作用机制。 Objective To evaluate the effect of small interference RNA(siRNA)histone deacetylase 6(HDAC6)on cisplatin sensitivity for cervical cancer cell Hela.Methods Cervical cancer cell Hela were chosen,and respectively transfected control siRNA and HDAC6 siRNA.Different concentrations of cisplatin were added to the transfection cells.The HDAC6 mRNA expression was detected by RT-PCR.The HDAC6,Bcl-2 and Bax protein expression was detected by Western blot.The cell apoptosis was detected by flow cytometry.The cell survival rate in vitro was detected by MTT method.Results The HDAC6 mRNA expression of HDAC6 siRNA group(82.3±24.5)were significantly lower than blank control group(352.7±89.2)and control siRNA group(361.7±77.6)(all P<0.05).The HDAC6 and Bcl-2 protein expression of HDAC6 siRNA group[(118.7±20.9),(179.5±44.6)]were significantly lower than blank control group[(558.4±105.5),(494.1±65.3)]and Control siRNA group[(564.9±98.5),(487.2±74.1)](all P<0.05).However,the Bax protein expression of HDAC6 siRNA group(448.6±70.8)were significantly higher than blank control group(189.7±60.3)and control siRNA group(194.2±68.4)(all P<0.05).The cell apoptosis rate of HDAC6 siRNA group(37.6±8.7)%was significantly higher than blank control group(5.8±1.1)%and control siRNA group(6.4±1.3)%(all P<0.05).After different concentration cisplatin treatment,the IC50 value of HDAC6 siRNA group significantly was lower than blank control group and control siRNA group(all P<0.05).Conclusion Silencing HDAC6 expression can improve the cisplatin sensitivity of cervical cancer cell Hela,and promoting cell apoptosis may be the main mechanism
