蝇蛆抗菌肽对小鼠金黄色葡萄球菌性乳腺炎治疗效果的研究 被引量:2

摘要 目的研究蝇蛆抗菌肽对小鼠金黄色葡萄球菌性乳腺炎的治疗效果。方法本试验采用金黄色葡萄球菌诱导蝇蛆,从蝇蛆中提取抗菌肽并用于治疗金黄色葡萄球菌性乳腺炎。采用分娩后6~8 d的BALB/C雌鼠120只,随机分组:空白对照组、生理盐水治疗组、抗生素治疗组和抗菌肽治疗组,每组30只。空白对照组不做处理,另外3组用金黄色葡萄球菌感染小鼠建立小鼠乳腺炎模型,建模成功后,各试验组使用相对应的药物连续治疗7 d,并于第3、5、7天每组分别断颈处死10只小鼠,观察小鼠乳腺病理组织及HE染色切片,测定小鼠乳腺组织中TNF-α含量和金黄色葡萄球菌数量。结果经过金黄色葡萄球菌诱导产生的抗菌肽浓度显著高于未诱导组(P<0.05)、对金黄色葡萄球菌具有较强的抑菌活性、对金黄色葡萄球菌的最小抑菌浓度为1.5 mg/L,蝇蛆抗菌肽可显著抑制小鼠乳腺组织中金黄色葡萄球菌的生长(P<0.05)、降低乳腺组织的损伤和极显著下调TNF-α的含量(P<0.01)。结论蝇蛆抗菌肽对小鼠金黄色葡萄球菌性乳腺炎治疗有一定治疗效果。 Objective To study the therapeutic effect of AMPs from maggot on S.aureus mastitis in mice. M ethods In this experiment, the AMPs were extracted from maggots which induced by S.aureus, it was used to treat staphylococcal mastitis. To chose the number of 120 BALB/C female mice after delivery 6 d to 8 d were randomly divided into 4 groups of 30 each. There were blank control group, normal saline treatment group, antibiotic treatment group, and AMPs treatment group. The blank control group was not treated, the other three groups were infected with S.aureus to establish a mouse mastitis model. After the model was successfully established, each group was treated with corresponding drugs for 7 d continuously, the 10 mice were killed in each group on the 3,5,7 d respectively. R esults To observe the pathological tissue, HE staining and determine the content of TNF-α and the number of S.aureus in mouse mammary glands.The results showed that the concentration of AMPs induced by S.aureus was significantly higher than the uninduced group(P<0.05). Which had strong antibacterial activity against S.aureus.The minimal inhibitory concentration of S.aureus was 1.5 mg/L, the AMPs of maggots could significantly inhibit the growth of S.aureus in mammary gland of mice(P<0.05). The injury of mammary tissue was reduced and the content of TNF-α was significantly decreased(P<0.01).C onclusion The therapeutic effects of antimicrobial peptides on staphylococcal mastitis in mice is useful.
