山楂原花青素改善肥胖小鼠糖脂代谢紊乱及脂肪肝的研究 被引量:16

摘要 为了探讨山楂原花青素提取物(Hawthorn proantho cyanidins, HPC)对高脂饮食诱导的肥胖小鼠糖脂代谢紊乱与脂肪肝的影响,将60只雌性C57BL/6J小鼠随机分为5组:溶剂对照组、高脂模型组、HpC低(50mg/kg)、中(100 mg/kg)、高(200 mg/kg)剂量组,每组12只。除溶剂对照组外4组喂高脂饲料,同时3个干预组动物每日灌胃给与不同剂量的HpC连续8周。以小鼠血清中总胆固醇(TC),甘油三酯(TG),低密度脂蛋白胆固醇(LDL-C),高密度脂蛋白胆固醇(HDL-C)含量和空腹血糖值(Glu)为检测指标,评估HpC对高脂诱导小鼠高血脂和高血糖的改善作用。并采集小鼠肝脏组织,石蜡切片,HE染色,进行组织学观察。结果表明,HpC可抑制高脂饮食导致的肥胖,与模型对照组相比,降低了肥胖小鼠的体质量与体脂含量,并改善肥胖小鼠肝脏变性程度。同时HpC可显著降低小鼠TC、TG、LDL-C和Glu值,不同程度地提高了HDL-C水平。试验证明HpC具有改善高脂诱导肥胖小鼠的血脂和血糖异常,降低脂肪肝的功效。 To investigate the effects of Hawthorn Proantho Cyanidins(HPC) on glucose and metabolism disorder and fatty liver in obese mice induced by high-fat diet. 60 female C57 BL/6 J mice were randomly divided into 5 groups: solvent control group, high fat model group, low HPC dose group(50 mg/kg), medium HPC dose group(100 mg/kg), and high HPC dose group(200 mg/kg), with 12 mice in each group. The four groups were fed with high-fat diet except for the solvent control group, and the three groups of animals were given daily intragastric administration of HPC. Body mass was recorded weekly for 8 weeks. Total cholesterol(TC), triglyceride(TG), low density lipoprotein cholesterol(LDL-C), high density lipoprotein cholesterol(HDL-C), and blood glucose level(Glu) of mice were used as indicators to evaluate the effect of HPC on the improvement of hyperlipidemia and abnormal blood sugar in mice induced by high fat. The fatty livers of mice were collected for HE staining to observe the degree of denaturation of the liver. The results showed that compared with the model control group, HPC significantly decreased the levels of TC, TG, LDL-C and Glu in mice, and increased the level of HDL-C in different degrees. At the same time, HPC reduced the body weight and body fat content of obese mice, and improved the degree of liver degeneration. Therefore, HPC has the effect of improving abnormal blood lipids and blood sugar, and reducing the degree of fatty liver of obese mice induced by high fat diet.
